【摘要】：目的對從廣州白云國際機場口岸入境的一例自蘇里南歸國發(fā)熱人員進行寨卡病毒實驗室檢測。方法采集發(fā)熱人員的血液和唾液樣本,同時使用兩種寨卡病毒實時熒光RT-PCR試劑進行寨卡病毒核酸檢測。RT-PCR擴增寨卡病毒部分基因片段,并進行序列測定和同源性比較�；跀U增片段構建系統(tǒng)進化樹,分析輸入性病例的來源。結果實時熒光RT-PCR結果顯示,該病例血清樣本寨卡病毒核酸弱陽性、唾液樣本寨卡病毒核酸陽性。RT-PCR擴增出預期大小約1.5Kb片段,測序結果表明該片段與巴西寨卡病毒株(GenBank號KX197250)相應序列的同源性為99%。系統(tǒng)進化樹顯示該病毒屬亞洲譜系。結論根據(jù)患者流行病學史、臨床表現(xiàn)和病例標本核酸檢測情況,確診該病例為廣東首例從蘇里南地區(qū)輸入性寨卡病例。
[Abstract]:Objective to detect Zika virus in a case of returning fever from Suriname in Guangzhou Baiyun International Airport. Methods Blood and saliva samples of febrile persons were collected and sequenced and homology were compared by using two kinds of real time fluorescent RT-PCR reagent to detect Zika virus nucleic acid and amplify partial DNA fragments of Zika virus by reverse transcription-polymerase chain reaction (RT-PCR). Phylogenetic tree was constructed based on amplified fragments and the origin of imported cases was analyzed. Results the results of real-time fluorescence RT-PCR showed that Zika virus nucleic acid was weakly positive in serum and saliva samples were positive for Zika virus nucleic acid. RT-PCR amplified the expected size of Zika virus 1.5Kb fragment. The sequencing results showed that the sequence of the fragment was 99% homology with the corresponding sequence of Brazilian Zika virus strain (GenBank KX197250). The phylogenetic tree shows that the virus belongs to the Asian lineage. Conclusion according to the epidemiological history, clinical manifestation and nucleic acid detection, the case was confirmed as the first imported Zika case from Suriname in Guangdong province.
