【摘要】:目的建立利用聚合酶螺旋反應(PSR)快速檢測甲型H1N1流感病毒的方法。方法針對甲型H1N1流感病毒的特異性HA基因設計了6套引物,通過實時濁度法和顯色法兩種方法判斷結(jié)果。結(jié)果與結(jié)論從6套引物中篩選出了最佳引物,并確定最佳溫度為65℃;進一步實驗表明最佳引物能特異性地檢測H1N1病毒,與14種其他呼吸道病原核酸無交叉反應,敏感性達到100拷貝,與PCR敏感性一致。所建立的方法簡單快速、特異性強、敏感性高,適合現(xiàn)場和基層單位應用推廣。
[Abstract]:Objective to establish a method for rapid detection of influenza A (H1N1) virus by polymerase helical reaction (PCR). Methods six sets of primers were designed for the specific HA gene of influenza A (H1N1) virus, and the results were judged by real-time turbidimetry and coloration.
