【摘要】：目的研究水痘-帶狀皰疹病毒(VZV)在人神經(jīng)母細胞瘤細胞株(SHSY5Y)中培養(yǎng)及增殖的特點,觀察VZV感染SHSY5Y細胞后的生物學效應。方法于普通倒置顯微鏡和電子顯微鏡下觀察被感染細胞的形態(tài)變化;利用CCK-8法檢測VZV感染對細胞增殖的影響;比較不同感染復數(shù)(MOI)和不同濃度胎牛血清維持液對病毒增殖的影響。結果普通倒置顯微鏡下觀察細胞病變效應(cytopathic effect,CPE),從24h~96h,細胞病變數(shù)量增加和病變程度加重。電鏡觀察顯示:感染48 h后核染色質固縮、濃染,線粒體肥大及增生,胞質空泡化和自噬小體形成,可見包裝成熟的病毒顆粒。CCK-8法提示VZV感染24h后細胞增殖被抑制,各實驗組存活率與對照組比較差異均有統(tǒng)計學意義(P0.05)。在2%胎牛血清的維持液中用MOI為1的VZV感染SHSY5Y細胞,能收獲較高滴度的病毒。結論初步建立了VZV感染SHSY5Y細胞的體外模型,并利用該模型觀察病毒感染對細胞生物性狀的影響,為探討VZV的致病及潛伏機制奠定了基礎。
[Abstract]:Objective to study the characteristics of the culture and proliferation of varicella-zoster virus (VZV) in human neuroblastoma cell line SHSY5Y, and to observe the biological effects of VZV infection on SHSY5Y cells. Methods the morphological changes of infected cells were observed under inverted microscope and electron microscope, and the effects of VZV infection on cell proliferation were detected by CCK-8 assay. The effects of different infective complex moi) and different concentrations of fetal bovine serum maintenance fluid on virus proliferation were compared. Results the cytopathic effect of cytopathic effect was observed under ordinary inverted microscope. From 24 h to 96 h, the number of cytopathic effect increased and the severity of cytopathic effect increased. Electron microscope observation showed that nuclear chromatin pyknosis, staining, hypertrophy and proliferation of mitochondria, vacuolation of cytoplasm and formation of autophagy were observed 48 h after infection. The survival rate of each experimental group was significantly different from that of the control group (P 0.05). In the maintenance solution of 2% fetal bovine serum, SHSY5Y cells were infected with VZV with MOI 1, and the higher titer of the virus was obtained. Conclusion the model of VZV infected SHSY5Y cells in vitro was established, and the effect of virus infection on cell biological characters was observed, which laid a foundation for the study of pathogenesis and latent mechanism of VZV.
