不同細菌所致小鼠血流感染模型中MIP-1β、MIP-2和IL-12p70的表達及變化規(guī)律

【摘要】:目的探討4種血流感染常見細菌致小鼠血流感染后MIP-1β、MIP-2和IL-12p70的表達及變化規(guī)律。方法建立金黃色葡萄球菌、糞腸球菌、大腸埃希菌和肺炎克雷伯菌CD-1(ICR)小鼠血流感染模型,采用Luminex液相芯片系統(tǒng)檢測各實驗組和PBS對照組小鼠感染后0.5、1、3、6、12、24和48 h MIP-1β、MIP-2和IL-12p70的含量。結果 MIP-1β的濃度在細菌入血后1 h時明顯升高,金黃色葡萄球菌、糞腸球菌、大腸埃希菌、肺炎克雷伯菌組和對照組濃度分別為(134.5±18.3)、(61.5±15.4)、(3 354.0±809.0)、(6 888.4±1 100.2)和(28.9±4.6)pg/mL;IL-12p70達峰值時的濃度分別為(389.3±118.1)、(127.6±10.0)、(42.2±3.5)、(62.8±8.4)和(4.8±0.3)pg/mL。MIP-1β、MIP-2的濃度在大腸埃希菌組和肺炎克雷伯菌組高于其他實驗組和對照組(均P0.01),而IL-12p70的濃度在金黃色葡萄球菌組和糞腸球菌組中高于大腸埃希菌組、肺炎克雷伯菌組和對照組(均P0.01)。結論大腸埃希菌組和肺炎克雷伯菌組MIP-1β和MIP-2的濃度高于金黃色葡萄球菌組和糞腸球菌組,而金黃色葡萄球菌組和糞腸球菌組IL-12p70的濃度高于大腸埃希菌組和肺炎克雷伯菌組。聯(lián)合檢測多種細胞因子或趨化因子在預測革蘭陽性或革蘭陰性菌感染方面有一定價值,且可能為感染早期指導治療提供依據。
[Abstract]:Objective to investigate the expression and changes of MIP-1 尾 -MIP-2 and IL-12p70 in mice after blood stream infection induced by four kinds of common bacteria. Methods the blood stream infection models of Staphylococcus aureus, Enterococcus faecalis, Escherichia coli and Klebsiella pneumoniae were established in mice. The levels of MIP-1 尾 -MIP-2 and IL-12p70 in each experimental group and PBS control group were detected by Luminex liquid chip system. Results the concentration of MIP-1 尾 was significantly increased at 1 hour after blood entry. Staphylococcus aureus, Enterococcus faecalis, Escherichia coli, and Escherichia coli were observed. The concentrations of IL-12p70 in Klebsiella pneumoniae group and control group were 134.5 鹵18.3 鹵15. 5 鹵15. 4, 3 354.0 鹵80. 0 0. 2 and 28. 9 鹵4. 6 mg / mL respectively. The peak concentrations of IL-12p70 were 389.3 鹵118.1 鹵127.6 鹵10. 5 鹵42. 8 鹵3. 8 鹵8. 4 and 4. 8 鹵0.3)pg/mL.MIP-1 尾 -MIP-2 in Escherichia coli and Klebsiella pneumoniae group, respectively (P 0. 01, P 0. 01), while those in IL-12p70 group were significantly higher than those in other experimental and control groups (P 0. 01, P 0. 01). The concentration in Staphylococcus aureus group and Enterococcus faecalis group was higher than that in Escherichia coli group. Klebsiella pneumoniae group and control group (P 0.01). Conclusion the concentrations of MIP-1 尾 and MIP-2 in Escherichia coli group and Klebsiella pneumoniae group were higher than those in Staphylococcus aureus group and Enterococcus faecalis group, while the IL-12p70 concentration in Staphylococcus aureus group and Enterococcus faecalis group was higher than that in Escherichia coli group and Klebsiella pneumoniae group. Combined detection of various cytokines or chemokines has certain value in predicting Gram-positive or Gram-negative bacteria infection, and may provide evidence for early treatment of infection.
