改良型TAT-PTD-VP3融合蛋白的基因合成、原核表達(dá)載體構(gòu)建及原核表達(dá)的實(shí)驗(yàn)研究

【摘要】:VP3是一種能特異性誘導(dǎo)惡性腫瘤細(xì)胞凋亡的病毒蛋白。由于它是一種不能透過(guò)細(xì)胞膜的大分子蛋白,,目前主要通過(guò)真核表達(dá)載體將其基因?qū)肽[瘤細(xì)胞內(nèi)表達(dá)而發(fā)揮作用。因轉(zhuǎn)基因操作困難、真核表達(dá)效率較低且其臨床應(yīng)用的安全性近年來(lái)受到了質(zhì)疑,因此如能在體外制備出VP3蛋白并通過(guò)有效的方法將其導(dǎo)入腫瘤細(xì)胞內(nèi)發(fā)揮作用,則能有效解決上述問(wèn)題。TAT-PTD是一種能快速高效的將分子量為15 X 10~3~2 X 10~5的大分子蛋白跨膜轉(zhuǎn)導(dǎo)入細(xì)胞內(nèi)的多肽,改良型TAT-PTD則是在TAT-PTD的基礎(chǔ)上改良而來(lái)的高效跨膜轉(zhuǎn)導(dǎo)多肽,其跨膜轉(zhuǎn)導(dǎo)大分子蛋白的效率是TAT-PTD的30多倍。因此如將上述兩者結(jié)合則可有效解決體外制備的VP3蛋白跨腫瘤細(xì)胞膜轉(zhuǎn)運(yùn)的問(wèn)題。 本論文就改良型TAT-PTD-VP3融合蛋白的基因合成、原核表達(dá)載體的構(gòu)建及其原核表達(dá)進(jìn)行了初步研究。首先通過(guò)基因拼接法合成VP3基因并委托生物技術(shù)公司合成改良型TAT-PTD基因。然后于原核表達(dá)載體pGEX-6p-1上構(gòu)建改良型TAT-PTD-VP3融合蛋白的基因。最后將構(gòu)建好的原核表達(dá)載體轉(zhuǎn)導(dǎo)入基因工程菌DH-5α中并誘導(dǎo)其表達(dá)。本研究成功合成了VP3基因,構(gòu)建了改良型TAT-PTD-VP3融合蛋白的原核表達(dá)載體,并經(jīng)基因測(cè)序證明構(gòu)建無(wú)誤;同時(shí)我們成功的在基因工程菌DH-5α中誘導(dǎo)了改良型TAT-PTD-VP3融合蛋白的表達(dá)
[Abstract]:VP3 is a viral protein that can specifically induce apoptosis of malignant tumor cells. Because it is a kind of macromolecular protein which can not penetrate the cell membrane, it mainly plays a role by introducing its gene into tumor cells by eukaryotic expression vector. Because of the difficulty of transgenic operation, the efficiency of eukaryotic expression is low and the safety of its clinical application has been questioned in recent years. Therefore, if VP3 protein can be prepared in vitro and introduced into tumor cells by effective methods, the above problems can be effectively solved.
