大鼠機械性腦損傷生物標志物的研究

【摘要】: 目的:研究閉合性顱腦損傷大鼠血清和海馬蛋白質(zhì)表達譜,探尋腦損傷生物標志蛋白。 方法:選雄性SD大鼠隨機分為手術對照組及損傷后4 h、8 h、12 h、24 h和48 h組,復制Marmarou’s落體打擊大鼠腦損傷模型。分別進行常規(guī)HE染色、尼氏染色;并用強陰離子交換芯片(SAX2)和疏水表面芯片(H4)結合表面增強激光解析電離飛行時間質(zhì)譜技術分析大鼠閉合性腦損傷后不同時間血清和海馬中蛋白質(zhì)表達譜的改變。 結果:1臨床表現(xiàn)損傷組大鼠于打擊后有84只大鼠(70%)出現(xiàn)抽搐同時有呼吸暫停,持續(xù)約15s;在沒有出現(xiàn)抽搐的大鼠中有11只大鼠(9.17%)出現(xiàn)驚厥,持續(xù)約30s;17只大鼠(14.3%)傷后有肢體活動受限,表現(xiàn)為單肢或單側肢體活動受限。12只大鼠(10%)于打擊后出現(xiàn)持續(xù)呼吸抑制和全身抽搐,幾十秒鐘后呼吸停止,死亡。 2大體病理損傷組大鼠可見蛛網(wǎng)膜下腔出血,分布廣泛,無明顯大面積挫傷或局灶性挫裂傷;切面可見胼胝體點狀出血及雙側腦室出血。 3光鏡下觀察損傷組大鼠HE染色主要表現(xiàn)為額葉、頂葉、枕葉、小腦及腦干蛛網(wǎng)膜下腔出血,側腦室出血,小挫傷灶。部分神經(jīng)細胞胞體濃縮,胞核固縮,胞漿內(nèi)尼氏小體消失呈同質(zhì)性嗜酸性著色(紅色神經(jīng)細胞);部分神經(jīng)細胞胞體腫脹,胞漿不規(guī)則淡染,核腫大淡染或消失。腦組織水腫,表現(xiàn)
[Abstract]:Aim: to study the protein expression profiles of serum and hippocampus in rats with closed brain injury and to explore the biomarkers of brain injury. Methods: male SD rats were randomly divided into two groups: the control group and the control group. The rats were randomly divided into two groups: the control group and the 24 h and 48 h groups after 4 h, 8 h and 12 h after injury. The model of brain injury was induced by Marmarou's falling body. Routine HE staining and Nissl staining were performed respectively. Strong anion exchange chip (SAX2) and hydrophobic surface chip (H4) combined with surface enhanced laser desorption ionization time of flight mass spectrometry were used to analyze the changes of protein expression profiles in rat serum and hippocampus at different time after closed brain injury. Results: 1 there were 84 (70%) rats (70%) with convulsions and apnea (15 s) in the clinical manifestation group, 11 (9.17%) of the rats (9.17%) had convulsions and 30 s of convulsions. 17 rats (14.3%) had limited limb movement after injury, which was characterized by limited movement of one or one limb, 12 rats (10%) had sustained respiratory inhibition and systemic convulsions after striking, and after dozens of seconds, breathing stopped and died. 2Subarachnoid hemorrhage was found in the gross pathological injury group, with no obvious large area contusion or focal contusion, and corpus callosum punctate hemorrhage and bilateral ventricular hemorrhage were observed on the cut surface. 3The HE staining of the injured rats under light microscope was mainly manifested as subarachnoid hemorrhage in frontal lobe, parietal lobe, occipital lobe, cerebellum and brainstem, lateral ventricular hemorrhage and small contusion. Some of the neuronal bodies were concentrated, the nuclei were constricted, and the Nissl bodies disappeared in the cytoplasm with homogeneous eosinophilic staining (red nerve cells), some of the neuronal cells were swollen, irregularly stained, and the nuclei were dilated or disappeared.
