姜黃素誘導腫瘤細胞自噬后的自噬體濃縮疫苗抗腫瘤的機制研究

【摘要】:目的研究姜黃素誘導腫瘤細胞自噬后的自噬體濃縮疫苗抗腫瘤的機制。方法細胞自噬通過Western blotting和透射電子顯微鏡。小鼠骨髓來源的樹突狀細胞(BMDCs)的成熟表型經(jīng)常規(guī)掃描電子顯微鏡(SEM)、流式細胞儀(FCM)和透射電子顯微鏡(TEM)證實。自噬體介導的交叉呈遞效率通過羥基熒光素二醋酸鹽琥珀酰亞胺脂(CFSE)的分布測定T細胞的增殖。分析自噬體的抗原交叉呈遞的機理。結果姜黃素誘導的自噬通過自噬體的電子顯微鏡以及LC3-Ⅱ的表達所證實。自噬體濃縮疫苗上調(diào)表達CD40、CD86和MHCⅡ。經(jīng)自噬體介導的T細胞激活后,T細胞有明顯的擴增同時能殺傷靶細胞。HSP90、HSP70和HMGB1在這種新的疫苗中發(fā)揮重要作用。結論這些數(shù)據(jù)表明以自噬體為基礎的疫苗有抗腫瘤療效獨特的特點。這種新的腫瘤疫苗的功效需臨床試驗的進一步驗證。
