賀斯在體外對健康人免疫系統(tǒng)細胞因子的影響

【摘要】:目的 探討不同濃度的羥乙基淀粉類血漿代用品(賀斯,6%HAES 200/0.5)對正常人體免疫系統(tǒng)白介素-2,10,12(IL-2,10,12)及干擾素-γ(IFN-γ)的影響,以檢驗其是否抑制正常人體的免疫系統(tǒng),為在圍術期替代輸血提供試驗依據(jù),以降低輸血造成的惡性腫瘤的復發(fā)率及術后感染率和病死率。 方法 正常志愿者12名,嚴格無菌條件下抽取靜脈血各20毫升,并以肝素抗凝,在超凈臺內(nèi)分離出單個核細胞,計數(shù)后將細胞平均分成5組:1640培養(yǎng)液陰性對照組、10%血漿對照組、10%賀斯試驗組、20%賀斯試驗組、40%賀斯試驗組,然后再將各組分成陰性對照組、加脂多糖(LPS)組、加植物血凝素(PHA)組,使單個核細胞在不同的培養(yǎng)液中培養(yǎng)48小時后,離心提取上清液,以酶聯(lián)免疫吸附法(ELISA法)檢測各上清液中細胞因子白介素-2,10,12(IL-2,IL-10,IL-12)和干擾素-γ(IFN-γ)的濃度。 結(jié)果 三個濃度的賀斯試驗組與1640培養(yǎng)液及血漿對照組在三種環(huán)境(陰性對照、加LPS、加PHA)中比較,雖然四種細胞因子的濃度值稍有參差,但統(tǒng)計學顯示無顯著性差異(p0.05);在IL-2和IFN-γ,陰性對照組與加LPS組無顯著性差異(p0.05),而兩者均與加PHA組有顯著性差異(p0.05);在IL-10,IL-12,陰性對照組、加LPS組及加PHA組之間均示有顯著性差異(p0.05)。 結(jié)論 三個濃度的賀斯下培養(yǎng)單個核細胞48小時后與同條件下的血漿組及陰性培養(yǎng)液組的細胞因子濃度無顯著差異,提示三個濃度的賀斯在體外環(huán)境中不影響免疫細胞的生存、克隆及細胞因子的產(chǎn)生,即:賀斯在體外試驗
[Abstract]:Objective to investigate the effects of different concentrations of hydroxyethyl starch plasma substitutes (HES 200 / 0.5) on the levels of interleukin-2siloxacin 1012 (IL-2m10N12) and interferon- 緯 (IFN- 緯) in normal human immune system in order to determine whether they inhibit the immune system of normal human beings. In order to reduce the recurrence rate, postoperative infection rate and mortality of malignant tumor caused by blood transfusion, we can provide experimental basis for replacing blood transfusion in perioperative period. Methods Mononuclear cells were isolated from 12 normal volunteers under strict aseptic conditions by extracting 20 ml of venous blood and using heparin as anticoagulant. After counting, the cells were divided into 5 groups, 10% plasma control group, 10% Horse test group and 40% Horse test group, then each group was divided into negative control group, lipopolysaccharide (LPS) group and phytohemagglutinin (PHA) group. After the mononuclear cells were cultured in different culture medium for 48 hours, the supernatants were extracted by centrifugation. The concentrations of cytokines interleukin-210-10 (IL-12) and interferon- 緯 (IFN- 緯) in the supernatants were detected by enzyme-linked immunosorbent assay (ELISA). Results the concentrations of four cytokines were slightly different between the three concentrations of Horse test group and 1640 medium and plasma control group in three environments (negative control, plus LPS, plus PHA), although the concentrations of four cytokines were slightly different. In IL-2 and IFN- 緯, there was no significant difference between the negative control group and the LPS group (p0.05), but there was a significant difference between the two groups (p0.05), and there was a significant difference between the IL-10 IL-12 group, the negative control group, the LPS group and the PHA group (p0.05). Conclusion there was no significant difference in cytokine concentrations between the three concentrations of Hos cultured mononuclear cells in vitro and plasma and negative medium groups after 48 hours, suggesting that the three concentrations of Hos did not affect the survival of immune cells in vitro.
