表皮葡萄球菌mscL基因突變株的構(gòu)建及其生物學(xué)功能

【摘要】:目的初步探討表皮葡萄球菌mscL基因的生物學(xué)功能。方法構(gòu)建含有壯觀霉素抗性基因(spc)的同源重組質(zhì)粒pMAD-ΔmscL,電轉(zhuǎn)入表皮葡萄球菌1457,在42℃條件下振蕩培養(yǎng),利用藍(lán)白斑和抗生素抗性篩選表皮葡萄球菌mscL基因敲除突變株(SE1457-ΔmscL)。通過檢測低滲脅迫前后突變株和野生株D600值及CFU的變化觀察mscL基因?qū)B透壓的調(diào)節(jié)作用。采用微量板半定量方法檢測不同條件下mscL敲除突變對細(xì)菌生物膜形成的影響。結(jié)果成功構(gòu)建同源重組質(zhì)粒pMAD-ΔmscL,經(jīng)PCR擴增和測序驗證獲得了表皮葡萄球菌mscL基因敲除突變株,并通過RT-PCR在轉(zhuǎn)錄水平上得到進(jìn)一步驗證。處于對數(shù)生長期的mscL突變株在低滲脅迫下的生存能力顯著降低,但其生物膜形成能力與野生株相比無顯著差異。結(jié)論表皮葡萄球菌mscL基因可在對數(shù)生長期參與滲透壓的調(diào)節(jié),但對生物膜的形成無影響。
