兔中性粒細胞防御素對A549細胞IL-8基因表達的影響

【摘要】:【背景】脊椎動物防御素(defensins)是小分子的陽離子抗菌多肽,它們廣泛分布于上皮細胞和吞噬細胞中。人和兔中性粒細胞的總蛋白中5%以上為防御素,它們除了具有直接的抗菌功能外還能作為免疫活性分子作用于上皮細胞和免疫細胞,參與天然免疫和獲得性免疫的調(diào)節(jié)。另外它們還可作為信號分子調(diào)節(jié)細胞信號轉(zhuǎn)導(dǎo)過程,,促進細胞的增殖、分化等。探索防御素的非抗菌功能是目前防御素研究的重點方向之一,本研究從mRNA和蛋白水平初步探討了兔中性粒細胞防御素(rabbits neutrophils antimicrobial peptides,NPs)對人Ⅱ型肺泡上皮細胞A549 IL-8表達的影響及其作用機制。 【方法與結(jié)果】向家兔腹腔注射1%酪蛋白350ml,構(gòu)建兔腹腔無菌性炎癥動物模型。收集腹腔炎性滲出液,離心后棄上清,用雙蒸水破紅細胞,離心后超聲破碎白細胞,再次離心收集上清。上清經(jīng)Bio-Gel P-10低壓色譜凝膠層析柱分離純化后,用RP-HPLC法經(jīng)C18柱進一步純化,

More than 5% of the total proteins in human and rabbit neutrophils are defensins. In addition to their direct antimicrobial function, they can act as immune active molecules in epithelial cells and immune cells, and participate in the regulation of innate and acquired immunity.
