【摘要】：目的:通過(guò)檢測(cè)磷酸化p38 MAPK(p-p38MAPK)在小鼠卵泡發(fā)育過(guò)程中的表達(dá),為研究絲裂原活化蛋白激酶(MAPKs)信號(hào)通路在卵泡生長(zhǎng)、發(fā)育及成熟過(guò)程中的作用提供理論基礎(chǔ)。方法:隨機(jī)選取健康成年小鼠60只,按雌雄2∶1的比例合籠繁殖新生小鼠。取3、5、7、11及21天雌性健康小鼠(各30只)的卵巢組織,采用免疫組化SP法檢測(cè)不同年齡段小鼠卵巢組織中p-p38MAPK蛋白的表達(dá)水平。結(jié)果:p-p38MAPK蛋白在各年齡段小鼠不同級(jí)別卵泡的卵母細(xì)胞胞漿中均有表達(dá),在始基卵泡中表達(dá)最強(qiáng),與各級(jí)卵泡相比,差異均有統(tǒng)計(jì)學(xué)意義(P0.05);初級(jí)卵泡與次級(jí)卵泡比較,差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05);初級(jí)卵泡、次級(jí)卵泡與竇狀卵泡相比,差異均有統(tǒng)計(jì)學(xué)意義(P0.05)。該蛋白在各級(jí)卵泡顆粒細(xì)胞中也均表達(dá),始基卵泡與各級(jí)卵泡相比,差異均有統(tǒng)計(jì)學(xué)意義(P0.05);初級(jí)卵泡與次級(jí)卵泡相比差異有統(tǒng)計(jì)學(xué)意義(P0.05),而與竇狀卵泡相比差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05);在次級(jí)卵泡中表達(dá)最強(qiáng),與各級(jí)卵泡相比差異均有統(tǒng)計(jì)學(xué)意義(P0.05)。不同年齡段小鼠卵巢中,21天小鼠卵母細(xì)胞p-p38MAPK蛋白表達(dá)強(qiáng)度分別與3天和7天小鼠進(jìn)行比較,差異均有統(tǒng)計(jì)學(xué)意義(P0.05);3、5、7及11天各組相比,差異均無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。各年齡組顆粒細(xì)胞p-p38MAPK蛋白表達(dá)強(qiáng)度相比,差異均有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論:p-p38MAPK蛋白在小鼠卵泡發(fā)育過(guò)程中具有時(shí)空特異性,p-p38MAPK通路可能在卵泡發(fā)育過(guò)程中起重要的調(diào)控作用。
[Abstract]:Objective: to provide a theoretical basis for the study of the role of mitogen activated protein kinase (MAPKs) signaling pathway in the process of follicle growth, development and maturation by detecting the expression of phosphorylated p38 MAPK (p-p38MAPK) in the follicular development of mice. Methods: 60 healthy adult mice were randomly selected to reproduce the newborn babies according to the proportion of male and female 2 to 1. The expression level of p-p38MAPK protein in ovarian tissue of mice of different ages was detected by immunohistochemical SP method. Results: the expression of p-p38MAPK protein was expressed in the cytoplasm of different levels of the oocytes of mice of different ages, and the expression of the p-p38MAPK protein was the strongest in the basal follicles. The difference between the primary follicles and the secondary follicles was statistically significant (P0.05). The primary follicle and the secondary follicle were not statistically significant (P0.05). The primary follicle and secondary follicle were significantly different from the sinus follicles (P0.05). The protein was also expressed in the follicle granulosa cells at all levels, and the basal follicles were different from the follicles at all levels. Statistical significance (P0.05); the primary follicle had a statistically significant difference compared with the secondary follicle (P0.05), but had no statistical significance compared with the sinus follicle (P0.05); the strongest expression in the secondary follicle was statistically significant (P0.05). In mice of different age groups, the 21 day mouse oocyte p-p38MAPK eggs The white expression intensity was compared with the 3 day and 7 day mice, the difference was statistically significant (P0.05), and there was no significant difference between 3,5,7 and 11 days (P0.05). The difference of p-p38MAPK protein expression intensity in all age groups was statistically significant (P0.05). Conclusion: p-p38MAPK protein is in the process of mouse follicle development. Sometimes the p-p38MAPK pathway may play an important role in the regulation of follicular development.
