透明質(zhì)酸對人骨髓來源間充質(zhì)干細(xì)胞軟骨分化的影響

【摘要】:目的:探討外源性透明質(zhì)酸(hyaluronic acid,HA)對人骨髓來源間充質(zhì)干細(xì)胞(Human bone marrow derived mesenchymal stemcells,h-Mscs)增殖、向成軟骨方向定向分化等生物學(xué)行為的影響及其機(jī)制,為Mscs構(gòu)建組織工程化軟骨奠定基礎(chǔ)。 方法:取人骨髓來源間充質(zhì)干細(xì)胞體外培養(yǎng),將傳代后的細(xì)胞分別置于含有0.1mg/ml和0.2mg/mlHA的誘導(dǎo)培養(yǎng)基中進(jìn)行培養(yǎng),將具有促進(jìn)細(xì)胞增殖分化及抑制多種炎性介質(zhì)活性等多重生物學(xué)效應(yīng)的TGFβ1以10ng/ml濃度配置無血清培養(yǎng)基誘導(dǎo)同組Mscs作陽性對照,觀察細(xì)胞的增殖,分化。用Ⅱ型膠原免疫組化染色評(píng)價(jià)不同誘導(dǎo)因素下的軟骨基質(zhì)合成情況。 結(jié)果:所有細(xì)胞顯示了良好的增殖能力,暴露于外源性透明質(zhì)酸下的Mscs保持了良好的生長活性,并開始向軟骨表型分化,與未添加誘導(dǎo)因子組細(xì)胞相比差異有顯著性意義。不同濃度HA組的細(xì)胞合成軟骨基質(zhì)的能力無差異,但均不如陽性對照組。 結(jié)論:外源性HA具有誘導(dǎo)體外培養(yǎng)的人Mscs向軟骨表型分化的能力,但與TGF β1的誘導(dǎo)能力相比有差距。提示HA不但是構(gòu)成軟骨基質(zhì)機(jī)械
[Abstract]:Objective: to investigate the effects of exogenous hyaluronic acid (hyaluronic acidoha) on the proliferation and directional differentiation of human bone marrow derived mesenchymal stem cells (hMSCs) and its mechanism, so as to lay a foundation for the construction of tissue-engineered cartilage by mesenchymal stem cells (MSCs). Methods: human bone marrow-derived mesenchymal stem cells were cultured in vitro and cultured in induction medium containing 0.1mg/ml and 0.2 mg / ml HA, respectively.
