【摘要】：目的:研究 EDS-40 玻璃化溶液凍貯小鼠及人卵母細(xì)胞的效果。 方法:設(shè)計(jì)研究 EDS-40 玻璃化溶液。按隨機(jī)分組原則,分為實(shí)驗(yàn)組和對照組,實(shí)驗(yàn)組(a)采用自行設(shè)計(jì)的 EDS-40 玻璃化液,對照組(b)采用國際上公認(rèn)的效果最好的 EFS-40 玻璃化溶液,對照組(c)采用程序冷凍法。 1、對 EDS-40 溶液行玻璃化測試。 2、鼠卵凍貯:將小鼠卵母細(xì)胞按上述原則隨機(jī)分成 a、b、c 三組, a、b 組在 25°C 室溫下,分別加入 EFS-40 和 EDS-40 玻璃化溶液后裝管并迅速投入液氮中,c 組用程序冷凍法進(jìn)行凍存。三組凍貯 2～ 3 個月后復(fù)溫,用培養(yǎng)液反復(fù)洗滌后移入培養(yǎng)孔板培養(yǎng),觀察卵母細(xì)胞的成活率。將成活的卵母細(xì)胞行體外受精,觀察其受精的情況。實(shí)驗(yàn)結(jié)果用 SPSS11.5 軟件進(jìn)行統(tǒng)計(jì)學(xué)分析。 3、人卵凍貯:在成功玻璃化冷凍小鼠卵母細(xì)胞的基礎(chǔ)上,再隨機(jī)選用人卵母細(xì)胞,其冷凍方法、復(fù)溫方法及統(tǒng)計(jì)學(xué)處理同動物實(shí)驗(yàn)。 結(jié)果 : 1、EDS-40 玻璃化溶液能達(dá)到玻璃化效果。 2、對小鼠卵母細(xì)胞兩種玻璃化溶液的毒性有顯著性差異,EDS-40較 EFS-40 毒性低。小鼠卵母細(xì)胞玻璃化冷凍后各組存活率分別為:79.34%、67.94%、56.34%,受精率分別為:79.17%、66.29%、51.14%。 3、人卵母細(xì)胞冷凍后 a、b、c 三組各組存活率分別為:53.57 %、66.67%、56.67 %,受精率為 40.00%、77.78%、41.18%。
[Abstract]:Objective: to study the effect of cryopreservation of mouse and human oocytes with EDS-40 vitrification solution. Methods: the glass solution of EDS-40 was designed and studied. According to the principle of random grouping, the experimental group and control group were divided into experimental group and control group. The experimental group was treated with self-designed EDS-40 vitrification solution, while the control group was treated with EFS-40 vitrification solution, which was internationally recognized as the best. (1) vitrification of EDS-40 solution. 2. Storage of mouse eggs. The oocytes were randomly divided into three groups according to the above principles. After adding EFS-40 and EDS-40 vitrified solution respectively, the tube was filled and rapidly put into liquid nitrogen group C for freezing by programmed freezing method. After 2 ~ 3 months of frozen storage, the three groups were incubated by repeated washing of culture medium and transferred into the culture hole plate to observe the survival rate of oocytes. The surviving oocytes were fertilized in vitro to observe their fertilization. The results were statistically analyzed by SPSS 11.5 software. 3. Human oocyte storage: on the basis of successful vitrification of mouse oocytes, human oocytes were selected randomly. The rewarming method and statistical treatment were the same as the animal experiment. Results: 1 the vitrification effect of EDS-40 was achieved. 2. Two kinds of vitrification solutions were applied to mouse oocytes. The toxicity of EDS-40 was lower than that of EFS-40. The survival rate of each group after vitrified freezing of mouse oocytes was 79.34 and 67.94 and 56.34, respectively. The fertilization rate was 79.1766.291.14.3, the survival rate of human oocyte frozen group was 53.57, 66.6756.67, and the fertilization rate was 40.000.77.780.The survival rate of each group was 41.180.The survival rate of each group was 53.57.
