肝前型門靜脈高壓癥大鼠模型建立的實(shí)驗(yàn)研究

【摘要】: 目的 本實(shí)驗(yàn)旨在通過部分結(jié)扎Wistar大鼠的門靜脈以建立穩(wěn)定的肝前型門靜脈高壓模型,并觀測本模型門靜脈壓力的變化,以利于對肝前型門靜脈高壓癥病理生理及治療方法進(jìn)一步研究提供動物模型。 材料和方法 取30只Wistar雄性大鼠作實(shí)驗(yàn)動物,平均體重300g(280-320g)。動物隨機(jī)分為兩組:A組大鼠10只,為假手術(shù)組(Sham組),開腹游離門靜脈后關(guān)腹;B組大鼠20只,為實(shí)驗(yàn)組(Portal vein ligation, PVL組),開腹用21G鈍針頭縮窄門靜脈后關(guān)腹,結(jié)扎前測量門靜脈直徑和門靜脈壓力,兩組術(shù)后2、6、10周測定門靜脈壓力,觀察腹腔內(nèi)門腔側(cè)枝循環(huán)形成情況。結(jié)果 術(shù)后1周內(nèi)大鼠體重下降,術(shù)后2周體重較術(shù)前增長。PVL組大鼠結(jié)扎術(shù)后2周、6周、10周門靜脈壓力(17.32±1.77cmH2O、17.36±1.93cmH2O、16.82±2.20 cmH2O)與結(jié)扎前壓力(9.21±0.63cmH2O)比較均有顯著性差異。Sham組大鼠術(shù)后2周、6周、10周門靜脈壓力(9.22±0.49cmH2O、9.27±0.43cmH2O、9.21±0.49cmH2O)與PVL組大鼠同期壓力比較有顯著性差異。本次實(shí)驗(yàn)大鼠門靜脈縮窄率約為88.9%。實(shí)驗(yàn)中發(fā)現(xiàn)大鼠腹腔出現(xiàn)了大量的門腔靜脈間側(cè)枝循環(huán),包括腹膜后門腔靜脈間的靜脈叢、左腎上腺靜脈明顯擴(kuò)張并肉眼可見與門靜脈所屬的曲張靜脈叢相連等。 結(jié)論 用21G鈍針和4號絲線對Wistar大鼠行門靜脈部分結(jié)扎術(shù),可以得到滿意的肝前型門靜脈高壓Wistar大鼠模型,本實(shí)驗(yàn)結(jié)果可以作為制作肝前型門靜脈高壓癥Wistar大鼠模型的標(biāo)準(zhǔn)。
[Abstract]:Objective to establish a stable prehepatic portal hypertension model by partial ligation of portal vein in Wistar rats and observe the changes of portal vein pressure in this model. Materials and methods 30 male Wistar rats were used as experimental animals with an average body weight of 300g / 320g / kg. The animals were randomly divided into two groups: sham group (n = 10), sham group (n = 20), portal vein ligation group (n = 20) and portal vein ligation group (n = 20). The diameter and pressure of portal vein were measured before ligation, and the portal vein pressure was measured 10 weeks after operation, and the formation of collateral circulation in abdominal cavity was observed. Results the weight of rats decreased within 1 week after operation. There were significant differences in body weight 2 weeks after operation compared with those before ligation. The portal vein pressure was 17.32 鹵1.77 cm H 2O 17.36 鹵1.93 cm H 2O 16.82 鹵2.20 cm H 2O at 2 weeks after ligation and 9.21 鹵0.63 cm H 2O before ligation. The portal vein pressure was 9.22 鹵0.49 cm H 2O 9.22 鹵0.49 cm H 2O 9.27 鹵0.43 cm H 2O 9.27 鹵0.49 cm H 2O 9.27 鹵0.43 cm H 2O 9.21 鹵0.49 cm H 2O in the sham group. There was a significant difference in the pressure in the same period. The coarctation rate of portal vein in this experiment was about 88. 9%. It was found that a large number of portal vena cava collateral circulation appeared in the abdominal cavity of rats, including the venous plexus between the retroperitoneal vena cava. The left adrenal vein was obviously dilated and connected with the variceal plexus attached to the portal vein with the naked eye. Conclusion the partial ligation of portal vein was performed on Wistar rats with 21G blunt needle and 4 ~ (th) silk thread. A satisfactory model of prehepatic portal hypertension in Wistar rats can be obtained. The results of this experiment can be used as the standard for making Wistar rat model of prehepatic portal hypertension.
