禽流感核酸疫苗免疫保護性研究

【摘要】:本論文由二部分組成(一)DNA疫苗與滅活疫苗混合免疫和單獨免疫不同的動物能提供抗H5N1亞型禽流感病毒的保護性研究(二)血凝素(hemagglutinin,HA)和神經(jīng)氨酸酶(neuraminidase,NA)DNA免疫BALB/c小鼠抗H9N2亞型禽流感病毒的保護性研究。 (一) 高致病性H5N1型禽流感病毒在1997年第一次突破種屬障礙傳給人,造成至少18例感染,6人死亡。禽流感病毒還仍然活躍在各國家禽市場,并影響世界的經(jīng)濟秩序和威脅到人類的生命安全。因此,必須預先接種疫苗誘導保護性免疫。本實驗分別以6—8周齡BALB/C小鼠和三黃雞為實驗動物,分四組免疫:第一組,全病毒滅活疫苗初免,HA-DNA加強:第二組HA-DNA初免,滅活疫苗加強;第三組滅活疫苗免疫二次,第四組HA-DNA免疫二次,間隔三周,加強免疫后三周,用致死量(20×LD_(50))的H5N1亞型禽流感病毒(A/Chichen/JiangSu/01/2002)攻擊小雞和小鼠,測定動物血清抗體變化、動物的存活率及攻毒后第三天小雞泄殖腔的排毒量和小鼠肺部的病毒殘存量。結果表明四種免疫方式能為不同的動物提供抗H5N1型禽流感病毒保護和交叉保護。(二) H9N2型禽流感病毒廣泛分布于禽類中,并且在香港和廣東地區(qū)的人類感染者體內(nèi)能夠分離得到。我們應當尋找有效的途徑控制H9N2型禽流感給人類帶來的危害。最近分離的A/Chichen/JiangSu/07/2002(H9N2)病毒經(jīng)肺對肺傳代能使BALB/C小鼠致死,表明H9N2型禽流感病毒能在小鼠肺部大量擴增。我們以BALB/C小鼠為動物模型,分別用3μg、10μg、30μgHA-DNA、NA-DNA電擊免疫6-8周齡的BALB/C小鼠一次或二次,一次免疫組于初免后四周用致死量(40×LD_(50))的H9N2型病毒攻擊。二次免疫組間隔三周后加強免疫,加強免疫后一周用致死量(40×LD_(50))
[Abstract]:This paper consists of two parts (one is mixed immunization with inactivated vaccine and the other is mixed immunization with inactivated vaccine) and the other is to immunize BALB/c mice with H5N1 subtype Avian Influenza virus.
