人牙髓干細(xì)胞的體外培養(yǎng)及神經(jīng)樣誘導(dǎo)分化

【摘要】:背景:牙髓組織來(lái)源干細(xì)胞的發(fā)現(xiàn)及概念的確立有助于從細(xì)胞水平上認(rèn)識(shí)牙齒發(fā)育和再生修復(fù)機(jī)制。目的:了解人牙髓干細(xì)胞向神經(jīng)元樣細(xì)胞誘導(dǎo)分化能力和誘導(dǎo)分化條件。方法:取健康青年人的第三磨牙的牙髓組織后,制成單細(xì)胞懸液,加入含有體積分?jǐn)?shù)為15%胎牛血清α-MEM培養(yǎng)基在6孔板培養(yǎng),傳代培養(yǎng)后加入丁羥基茴香醚、forskolin、β-巰基乙醇、堿性成纖維細(xì)胞生長(zhǎng)因子誘導(dǎo)劑進(jìn)行培養(yǎng)。結(jié)果與結(jié)論:免疫熒光和反轉(zhuǎn)錄-聚合酶鏈反應(yīng)檢測(cè)顯示,誘導(dǎo)2周后人牙髓細(xì)胞除了stro-1,Col-Ⅰ,牙本質(zhì)涎蛋白表達(dá)陽(yáng)性外,還有巢蛋白,神經(jīng)元特異性烯醇化酶的表達(dá)也是陽(yáng)性,而牙齦纖維細(xì)胞表達(dá)均為陰性。說(shuō)明人牙髓組織中存在成體干細(xì)胞,在一定的誘導(dǎo)培養(yǎng)條件下,牙髓干細(xì)胞有向神經(jīng)元樣細(xì)胞分化的潛能。
[Abstract]:Background: the discovery of dental pulp tissue derived stem cells and the establishment of their concepts are helpful to understand the mechanism of tooth development and regeneration at the cellular level. Objective: to understand the ability and inducement of human dental pulp stem cells to differentiate into neuron-like cells. Methods: the pulp tissue of the third molar of healthy young people was taken. Single cell suspension was prepared, and 偽 -MEM medium containing 15% fetal bovine serum was added in 6-well plate culture medium.
