不同電性脂質(zhì)體對金黃色葡萄球菌組氨酸激酶AgrC跨膜鑲嵌效率的影響

【摘要】:通過改變脂質(zhì)體中磷脂成分,構(gòu)建了不同電性的脂質(zhì)體.利用表面活性劑介導(dǎo)方法,將截短的金黃色葡萄球菌細(xì)胞膜上的組氨酸激酶AgrC(AgrCTM6-7C)蛋白重構(gòu)到不同電性的脂質(zhì)體上.結(jié)果表明,陰離子脂質(zhì)體對AgrCTM6-7C蛋白的鑲嵌效率明顯高于陽離子脂質(zhì)體,約60%~70%鑲嵌至陰離子脂質(zhì)體中的AgrCTM6-7C蛋白的細(xì)胞質(zhì)域朝向脂質(zhì)體囊泡的外部,并保持較高活性.利用圓二色光譜比較了AgrCTM6-7C蛋白在表面活性劑膠束和脂質(zhì)體中的二級結(jié)構(gòu)穩(wěn)定性,發(fā)現(xiàn)陰離子脂質(zhì)體對AgrCTM6-7C蛋白的二級結(jié)構(gòu)具有一定的保護(hù)作用,可明顯提高蛋白的熱穩(wěn)定性.
[Abstract]:Different electrical liposomes were constructed by changing the phospholipid composition of liposomes. The short staphylococcus aureus membrane histidine kinase AgrCon AgrCTM6-7C protein was reconstructed into different electrical liposomes. The inlay efficiency of anionic liposomes to AgrCTM6-7C protein was significantly higher than that of cationic liposomes. About 70% of the cytoplasmic domain of the AgrCTM6-7C protein embedded in the anion liposome is oriented towards the outside of the liposome vesicle. The secondary structure stability of AgrCTM6-7C protein in surfactant micelles and liposomes was compared by circular dichroism spectroscopy. It was found that anionic liposomes had a certain protective effect on the secondary structure of AgrCTM6-7C protein and could obviously improve the thermal stability of the protein.
