【摘要】:本實驗研究了胚胎期Wistar大鼠端腦及脊髓發(fā)育的基本過程。結(jié)果顯示E13天胎鼠神經(jīng)管已閉合,形成前端腦泡。此期前端腦泡尚未分化成端腦(見附圖1),腦泡上皮仍處于神經(jīng)管上皮階段;脊髓橫斷面呈實質(zhì)塊狀,神經(jīng)細胞排列密集,嗜堿性較強,未見到神經(jīng)纖維形成。至E15天,端腦開始發(fā)育,古皮質(zhì)形成,紋狀體原基形成;此期的脊髓橫斷面呈栗子狀,背側(cè)神經(jīng)細胞發(fā)育較快,HE染色嗜堿性較強,仍未見到神經(jīng)纖維形成。E17天始,新皮質(zhì)形成;脊髓灰質(zhì)橫斷面已漸呈蝴蝶結(jié)狀,背側(cè)雙角神經(jīng)元發(fā)育較前角快,細胞較小,HE染色嗜堿性較強;中央管橫斷面呈菱形;神經(jīng)纖維已清晰可見;在前角內(nèi)可見散在的尼氏體陽性細胞,胞體較大,多呈菱形。E19至E21天,端腦及脊髓的輪廓均已呈成年時形狀,端腦皮質(zhì)細胞分層漸清晰,脊髓前角內(nèi)神經(jīng)元清晰可見,神經(jīng)元胞體較大。同時本實驗還觀察了神經(jīng)元的發(fā)育情況。尼氏體于E15天始檢測到,但數(shù)量極少;E17天后數(shù)量逐漸增加,直至E19天。E13天至E15天(少量)雖未檢測到尼氏體,但此期的細胞小而圓,排列密集,嗜堿性強,說明此期是神經(jīng)元或神經(jīng)前體細胞的大量增殖期,而E19天后,至E21天,未見到神經(jīng)元數(shù)量的明顯增加,但胞體增大明顯,這說明此期是神經(jīng)元成熟期。 本實驗在觀察了大鼠胚胎時期端腦及脊髓的發(fā)育過程的基礎上,利用常規(guī)免疫組織化學方法探討了神經(jīng)生長因子特異性受體TrkA在胚胎發(fā)育不同時期端腦及脊髓內(nèi)的表達情況。結(jié)果發(fā)現(xiàn)E13天時胎鼠端腦及脊髓內(nèi)均未見到TrkA陽性表達,此時Nestin染色呈弱陽性,說明此階段處于神經(jīng)管發(fā)育的后期,還未形成特征性
[Abstract]:The basic process of the development of the terminal brain and spinal cord in embryonic Wistar rats was studied. The results showed that the neural tube of embryonic rat was closed on day 13. The anterior vesicles had not been differentiated into telencephalon at this stage (see Fig. 1), and the vesicular epithelium was still in the neurotubule epithelium stage. The transverse section of the spinal cord was parenchymal block, the nerve cells were dense and basophilic, and no nerve fibers were formed. On the 15th day of E15, the telencephalon began to develop, the paleocortex formed and the striatum primordium formed. The spinal cord in this stage was chestnut shaped, and the dorsal nerve cells developed faster and stronger basophilic with HE staining. However, the formation of nerve fibers and neocortex was not seen on the 17th day after the formation of nerve fibers. The cross section of the gray matter of the spinal cord had gradually appeared as a bow knot, the dorsal biangular neurons developed faster than the anterior horn, and the cells were smaller and stronger basophilic in HE staining. The cross section of the central tube was rhomboid. Nerve fibers were clearly visible. Scattered Nissl body positive cells were found in the anterior horn. The cell bodies were larger, most of them were rhombic. E19 to E21 days. The contours of the terminal brain and spinal cord were in the shape of adulthood, and the stratification of the terminal cortical cells was gradually clear. The neurons in the anterior horn of the spinal cord were clearly visible and the neuronal bodies were large. The development of neurons was also observed in this experiment. The Nissl's body was detected at the 15th day of E15, but the number was very small. After 17 days of E17, the number of cells increased gradually, until days E19, E13 to E15 (a little), although no Nieldite bodies were detected, the cells in this phase were small and round, densely arranged and strongly basophilic. The results showed that the proliferation of neurons or neural precursor cells was significant in this period, but no significant increase in the number of neurons was observed after E19 days and at 21 days after E21, but the increase of cell body was obvious. This indicates that this period is the mature stage of neurons. This experiment was based on the observation of the development of cerebrum and spinal cord in embryonic rats. The expression of nerve growth factor specific receptor (TrkA) in the telencephalon and spinal cord of fetal rats at different stages of embryonic development was studied by routine immunohistochemical method. The results showed that there was no expression of NGF-specific receptor TrkA in the telencephalon and spinal cord of fetal rats at day 13. Positive expression of TrkA was observed. At this time, Nestin staining was weakly positive, indicating that this stage was in the late stage of neural tube development and had not yet formed a characteristic.
