斑馬魚腦出血模型的建立及丹參在該模型上改善腦出血的研究

【摘要】:目的:建立阿托伐他汀誘導的斑馬魚腦出血模型,評價丹參煎液及其水溶性單體成分對斑馬魚腦出血的影響。方法:選用轉(zhuǎn)基因斑馬魚Tg(fli-1:EGFP)and Tg(gata1:DsRed)胚胎為研究載體,用阿托伐他汀誘導斑馬魚腦出血,建立腦出血斑馬魚模型,干預不同濃度的丹參煎液、丹酚酸A、丹酚酸B、丹參素,在熒光顯微鏡下拍照觀察,計算斑馬魚腦出血面積和出血率。結(jié)果:丹參煎液、丹酚酸A、丹酚酸B、丹參素均可減少斑馬魚腦出血面積,丹參煎液的作用最為明顯。而丹酚酸B不僅能減少出血面積,同時能夠降低出血率。結(jié)論:丹參其煎液和部分水溶性成分丹酚酸A、丹酚酸B、丹參素均對斑馬魚腦出血有改善作用。其中丹酚酸B改善斑馬魚腦出血的作用較為明顯,值得進一步研究。
[Abstract]:Aim: to establish a model of zebra fish intracerebral hemorrhage (ICH) induced by Atto vastatin and evaluate the effect of Salvia miltiorrhiza decoction and its water soluble monomers on ICH of zebrafish. Methods: transgenic zebrafish Tg (fli-1:EGFP) and Tg (gata1:DsRed) embryos were used to induce intracerebral hemorrhage (ICH) of zebra fish induced by Atto vastatin. The model of intracerebral hemorrhage was established to interfere with different concentrations of Salvia miltiorrhiza decoction and Salvianolic acid (SA). Salvianolic acid B and Danshensu were photographed and observed under fluorescence microscope to calculate the cerebral hemorrhage area and bleeding rate of zebra fish. Results: salvia miltiorrhiza decoction, Salvianolic acid A, Salvianolic acid B and Danshensu could reduce the area of cerebral hemorrhage of zebra fish, and the effect of Salvia miltiorrhiza decoction was the most obvious. Salvianolic acid B can not only reduce the bleeding area, but also reduce the bleeding rate. Conclusion: Salvianolic acid A, Salvianolic acid B and Danshensu can improve cerebral hemorrhage of zebra fish. The effect of Salvianolic acid B on improving cerebral hemorrhage of zebrafish is obvious, which is worthy of further study.
