【摘要】：目的檢測并比較Rv1789刺激結核分枝桿菌(Mtb)感染人群和健康對照者外周血IFN-γ水平。方法以原核表達體系構建、表達、純化rRv1789。從南京市疾病預防控制中心選擇39位受試者,參考臨床結核病診斷標準和rCM-WBIA診斷標準將其分為TB感染組(包括結核病患者和潛伏感染者)和健康對照組,進行rRv1789-WBIA試驗。結果成功構建pET30b-Rv1789質粒,SDS-PAGE鑒定表達產(chǎn)物分子質量單位為44ku,與預期相符;Western blot證實該蛋白能被相應抗體識別。rRv1789刺激Mtb感染組產(chǎn)生的IFN-γ水平顯著高于健康對照組(P0.01);rCM和rRv1789刺激受檢者全血IFN-γ水平具有顯著相關性(r=0.616,P0.01)。結論 PE/PPE家族蛋白Rv1789具有抗原性,可用作Mtb感染快速診斷和結核疫苗候選抗原。
[Abstract]:Objective to detect and compare the levels of IFN- 緯 in peripheral blood of Rv1789 stimulated Mycobacterium tuberculosis (Mtb) infected population and healthy controls. Methods rRv1789. was constructed, expressed and purified by prokaryotic expression system. Thirty nine subjects were selected from Nanjing Centers for Disease Control and Prevention. They were divided into TB infection group (including tuberculosis patients and latent infections) and healthy control group according to the diagnostic criteria of clinical tuberculosis and rCM-WBIA. The rRv1789-WBIA test was carried out. Results the pET30b-Rv1789 plasmid was successfully constructed, and the molecular weight unit of the expressed product was 44ku. which was consistent with the expectation. Western blot confirmed that the protein could be recognized by the corresponding antibody. The level of IFN- 緯 in Mtb infected group stimulated by rRv1789 was significantly higher than that in healthy control group (P0.01). There was a significant correlation between rCM and rRv1789 stimulated whole blood IFN- 緯 levels (r = 0.616, P 0.01). Conclusion PE/PPE family protein Rv1789 has antigenicity and can be used for rapid diagnosis of Mtb infection and candidate antigen for tuberculosis vaccine.
