【摘要】：蛋白質(zhì)棕櫚�；侵革柡褪嫉淖貦八猁}通過硫酯鍵或者酰胺鍵連接在蛋白質(zhì)肽鏈的半胱氨酸上,屬翻譯后修飾,可影響蛋白質(zhì)的相互作用、穩(wěn)定性及定位等功能。熱休克蛋白90(heat shock protein 90,Hsp90)是一種重要的分子伴侶,已證明其參與精子獲能等生理過程。然而,哺乳動(dòng)物精子中是否存在蛋白質(zhì)棕櫚�；�,Hsp90在精子不同生理狀態(tài)下是否發(fā)生棕櫚�；�,目前尚不清楚。本研究首先采用�；�-生物素置換法檢測小鼠附睪尾部成熟精子總蛋白質(zhì)棕櫚�；闆r,然后通過CSS-Palm 4.0軟件預(yù)測Hsp90的棕櫚�；稽c(diǎn),再進(jìn)一步結(jié)合免疫沉淀方法檢測附睪頭部、附睪尾部精子的Hsp90棕櫚�；闆r。結(jié)果顯示,小鼠附睪尾部精子多種蛋白質(zhì)存在棕櫚�；�,其中分子量大小約50、65、72、85和130 k Da的蛋白質(zhì)發(fā)生棕櫚�；顬轱@著;軟件預(yù)測顯示Hsp90兩個(gè)亞型共有5個(gè)棕櫚�；稽c(diǎn);免疫沉淀結(jié)果也顯示小鼠精子存在棕櫚�；腍sp90,且其棕櫚�；脚c小鼠精子的生理狀態(tài)有關(guān):附睪尾部棕櫚�；奖雀讲G頭部高,而獲能后的棕櫚�；奖全@能前明顯升高。以上結(jié)果表明,哺乳動(dòng)物精子中存在蛋白棕櫚�；�,且Hsp90棕櫚�；赡軈⑴c精子生理狀態(tài)的調(diào)節(jié)。
[Abstract]:Protein palmitoacylation means that the hexadecyl palmitate is connected to the cysteine of the protein peptide by thioester bond or amide bond. It is a post-translational modification which can affect the interaction, stability and localization of protein. Heat shock protein 90 (Hsp90) is an important molecular chaperone, which has been proved to be involved in physiological processes such as sperm capacitation. However, it is not clear whether there is protein palmitylation in mammalian sperm and whether Hsp90 occurs in different physiological state of sperm. In this study, the total protein palmitylation of mouse epididymal tail mature spermatozoa was detected by acyl-biotin replacement method, and then the CSS-Palm 4.0 software was used to predict the palmitoacylation site of Hsp90. The Hsp90 palmitylation of epididymal head and tail sperm was detected by immunoprecipitation. The results showed that there was palmitylation of many proteins in mouse epididymal tail sperm, among which the proteins with molecular weight of 5065kmol / 72O85 and 130k Da were the most significant, and the software prediction showed that the two subtypes of Hsp90 shared five palmitoacylation sites. The results of immunoprecipitation also showed that there was palmitoacylated Hsp90, in mouse spermatozoa and the level of palmitoacylation was related to the physiological state of mouse spermatozoa: the level of palmitoacylation in tail of epididymis was higher than that in head of epididymis. The level of palmitoacylation after capacitation was significantly higher than that before capacitation. These results suggest that there is protein palmitylation in mammalian spermatozoa and that Hsp90 palmitylation may be involved in the regulation of the physiological state of spermatozoa.
