小鼠舌菌狀乳頭及味蕾發(fā)育過(guò)程中組織形態(tài)變化及Shh的表達(dá)

【摘要】:背景:哺乳動(dòng)物的舌背黏膜表面與味覺(jué)相關(guān)的主要是菌狀乳頭和輪廓乳頭。味覺(jué)異常嚴(yán)重影響人們的生活質(zhì)量,因此了解菌狀乳頭及其味蕾的發(fā)育過(guò)程,對(duì)于解決因其異常導(dǎo)致的味覺(jué)減退有重要意義。目的:觀察小鼠舌菌狀乳頭及味蕾的發(fā)生、發(fā)育的組織形態(tài)變化。方法:取C57BL/6J小鼠胚胎期13.5-18.5 d(E13.5-E18.5),出生后0.5 d(PN0.5),PN4.5,PN7.5,PN14.5,PN21.5,PN90.5及出生后1.5年(PN1.5Y)的舌標(biāo)本,進(jìn)行掃描電鏡觀察及蘇木精-伊紅染色觀察,同時(shí)使用Shh抗體進(jìn)行免疫組織化學(xué)染色。取E16.5和E18.5小鼠舌標(biāo)本進(jìn)行S-100的免疫組織化學(xué)染色。結(jié)果與結(jié)論:(1)菌狀乳頭數(shù)量的決定主要發(fā)生于E13.5-E14.5。從E15.5開(kāi)始,菌狀乳頭形態(tài)上逐漸發(fā)育成熟。味蕾發(fā)育開(kāi)始于E16.5,至PN7.5味孔開(kāi)始形成。成年期和老年期味蕾細(xì)胞和細(xì)胞間質(zhì)減少,并可見(jiàn)到發(fā)生于菌狀乳頭周圍的角化;(2)Shh在E14.5表達(dá)于菌狀乳頭的上皮細(xì)胞,E16.5-E18.5表達(dá)于菌狀乳頭頂端的上皮細(xì)胞,并且在成年期和老年期的味蕾細(xì)胞都有所表達(dá)。說(shuō)明Shh在菌狀乳頭和味蕾發(fā)生、發(fā)育、成熟及維持的過(guò)程中發(fā)揮著重要作用;(3)在E18.5,S-100表達(dá)于菌狀乳頭下方的結(jié)締組織,說(shuō)明神經(jīng)長(zhǎng)入?yún)⑴c了味蕾發(fā)育完善成熟的過(guò)程。
[Abstract]:Background: the mucosal surface of the dorsal tongue of mammals is mainly associated with the taste of bacillary papilla and outline papilla. Taste abnormality seriously affects people's quality of life, so it is important to understand the development process of mycelial nipples and taste buds in order to solve the hypothyroidism caused by abnormal taste nipples. Objective: to observe the development and histological changes of tongue papilla and taste buds in mice. Methods: the tongue specimens of C57BL/6J mice were collected from embryonic stage 13.5-18.5 days (E13.5-E18.5), postnatal 0.5 day (PN0.5) PN4.5 and PN14.5PN21.5PN90.5 and 1.5 years postnatal (PN1.5Y). The samples were observed by scanning electron microscope and hematoxylin eosin staining. Meanwhile, Shh antibody was used for immunohistochemical staining. The tongue specimens of E16.5 and E18.5 mice were stained with S-100 immunohistochemical staining. Results and conclusions: (1) the determination of the number of bacteriform papilla mainly occurred in E13.5-E14.5. From E15. 5, the mycelial papilla developed and matured in shape. Taste buds began to develop in E 16. 5 and began to form taste holes in PN7.5. In adult and old age, taste bud cells and interstitial cells decreased, and keratosis occurred around the mycelial papilla. (2) Shh was expressed in the epithelial cells of E14.5-E18.5 at the apical end of the myceliform papilla. And both in adulthood and in old age, taste bud cells are expressed. It is suggested that Shh plays an important role in the development, maturation and maintenance of the papilla and taste buds. (3) the connective tissue under the myceliform papilla expressed in E18.5T S-100, which indicates that the nerve growth takes part in the process of taste bud development and maturation.
