復(fù)方星夏湯對(duì)痰濕型多囊卵巢綜合征模型小鼠腫瘤壞死因子-α、白細(xì)胞介素-6的影響

【摘要】:目的:觀察復(fù)方星夏湯對(duì)痰濕型多囊卵巢綜合征(PCOS)模型小鼠腫瘤壞死因子-α(TNF-α)、白細(xì)胞介素-6(IL-6)水平的影響,初步探究復(fù)方星夏湯治療痰濕型PCOS的作用機(jī)理。方法:采用硫酸脫氫表雄酮聯(lián)合鏈脲佐菌素及高脂高糖飼料法建立痰濕型PCOS小鼠模型。選取23日齡ICR雌性小鼠72只,隨機(jī)分為4組:空白對(duì)照組、模型組、復(fù)方星夏湯組、達(dá)英-35組,每組18只。比較各組小鼠體質(zhì)量變化、E2、T、LH、FSH、FPG、FINS、TNF-α、IL-6水平,并計(jì)算LH/FSH比值、HOMA-IR指數(shù),對(duì)所取卵巢進(jìn)行病理切片,在光學(xué)顯微鏡下觀察其組織形態(tài)學(xué)變化。結(jié)果:復(fù)方星夏湯組、達(dá)英-35組均可以使痰濕型PCOS小鼠體質(zhì)量,LH、LH/FSH比值,FPG、FINS、HOMA-IR指數(shù),TNF-α水平下降,保持或恢復(fù)卵巢正常組織形態(tài)結(jié)構(gòu);其中在降低IL-6水平、HOMA-IR指數(shù)方面顯著優(yōu)于達(dá)英-35組(P0.01)。結(jié)論:復(fù)方星夏湯可能通過降低血清中TNF-α、IL-6水平改善胰島素抵抗(IR),直接或間接調(diào)節(jié)內(nèi)分泌激素及生殖軸,促使卵泡發(fā)育成熟及排卵,提高生殖能力。
[Abstract]:Objective: to observe the effect of compound Xingxia decoction on the levels of tumor necrosis factor- 偽 (TNF- 偽) and interleukin-6 (IL-6) in (PCOS) model mice with phlegm-dampness type polycystic ovary syndrome (PCOS), and to explore the mechanism of Fufang Xingxia decoction in treating PCOS with phlegm dampness type. Methods: PCOS mice model of phlegm dampness was established by dehydroepiandrosterone sulfate combined with streptozotocin and high fat and high sugar diet. Seventy-two ICR female mice aged 23 days were randomly divided into 4 groups: blank control group, model group, compound Xingxia decoction group, Daying 35 group, 18 rats in each group. The body mass changes of each group were compared. The LH/FSH ratio and HOMA-IR index were calculated. The histological changes of the ovary were observed under optical microscope. Results: both the compound Xingxia decoction group and Da-35 group could decrease the level of TNF- 偽 and maintain or restore the normal structure of ovary in PCOS mice with phlegm-dampness type and the ratio of LHH / FSH to FINSHOMA-IR was significantly higher than that of Dain-35 group in decreasing the level of IL-6 (P0.01). Conclusion: compound Xingxia decoction can improve insulin resistance (IR), directly or indirectly regulate endocrine hormone and reproductive axis, promote follicle development, maturation and ovulation, and improve reproductive ability by reducing serum TNF- 偽 IL-6 level.
