健脾益氣攝血方對(duì)辛伐他汀誘導(dǎo)斑馬魚(yú)出血模型的止血效果研究

【摘要】:目的:觀察健脾益氣攝血方對(duì)辛伐他汀誘導(dǎo)斑馬魚(yú)出血的止血效果并與清熱涼血方比較。方法:用辛伐他汀0.5μmol/L處理AB系斑馬魚(yú)24 h,建立斑馬魚(yú)出血模型,在嚴(yán)格盲態(tài)實(shí)驗(yàn)條件下,在最大非致死量范圍內(nèi),應(yīng)用不同濃度實(shí)驗(yàn)藥物處理出血斑馬魚(yú),以出血減少率、止血率為主要觀察指標(biāo),以類似脾氣虛特點(diǎn)的血流量、血流量改善率與運(yùn)動(dòng)速度、運(yùn)動(dòng)改善率為附加指標(biāo),綜合評(píng)價(jià)健脾益氣攝血方的干預(yù)效果。結(jié)果:實(shí)驗(yàn)藥物B1在濃度為500和1 000μg/ml時(shí)出血斑馬魚(yú)的出血率分別為30%和15%,止血率分別為60%和80%;實(shí)驗(yàn)藥物B2在濃度為500和1 000μg/ml時(shí)出血斑馬魚(yú)出血率分別為45%和40%,止血率分別為40%和47%。表明在降低斑馬魚(yú)出血率與止血效果兩方面實(shí)驗(yàn)藥物B1優(yōu)于實(shí)驗(yàn)藥物B2。在相等濃度下,在提高出血斑馬魚(yú)血流量與血流量改善率兩方面實(shí)驗(yàn)藥物B1優(yōu)于實(shí)驗(yàn)藥物B2。在相等濃度下,在促進(jìn)斑馬魚(yú)運(yùn)動(dòng)速度與提高斑馬魚(yú)運(yùn)動(dòng)改善率方面實(shí)驗(yàn)藥物B1優(yōu)于實(shí)驗(yàn)藥物B2。結(jié)論:健脾益氣攝血方對(duì)辛伐他汀誘導(dǎo)的斑馬魚(yú)出血有良好的止血效果,并能夠改善出血斑馬魚(yú)血流量與提高運(yùn)動(dòng)速度,為健脾益氣攝血方治療脾不統(tǒng)血證提供了實(shí)驗(yàn)依據(jù)。
[Abstract]:Aim: to observe the hemostatic effect of Jianpi Yiqi decoction on zebrafish bleeding induced by simvastatin and compare it with Qingre Liangxue recipe. Methods: AB zebrafish were treated with simvastatin 0.5 渭 mol/L for 24 h. Under strict blind experimental conditions, different concentrations of experimental drugs were used to treat the bleeding zebrafish in the range of maximum non-lethal dose, in order to reduce the bleeding rate. The hemostatic rate was regarded as the main observation index, with the blood flow, the improvement rate of blood flow, the speed of movement and the rate of improvement of exercise as the additional indexes, the intervention effect of the prescription for invigorating spleen and invigorating qi was evaluated synthetically. Results: the bleeding rate of zebrafish was 30% and 15.5%, the hemostatic rate was 60% and 80%, respectively, when the concentration of B1 was 500 渭 g/ml and 1 000 渭 g/ml, the bleeding rate of zebrafish was 45% and 40% when the concentration of drug B2 was 500 渭 g/ml and 1 000 渭 g/ml, respectively. The blood rates were 40% and 47%, respectively. The results showed that the experimental drug B1 was superior to the experimental drug B2 in reducing the bleeding rate and hemostatic effect of zebrafish. At the same concentration, the experimental drug B1 was superior to the experimental drug B2 in increasing the blood flow and improving the blood flow rate of zebrafish. At the same concentration, the experimental drug B1 was superior to the experimental drug B2 in promoting the speed of zebrafish movement and improving the rate of zebrafish movement improvement. Conclusion: Jianpi Yiqi decoction has good hemostatic effect on zebrafish bleeding induced by simvastatin and can improve the blood flow and speed of movement of zebrafish.
