【摘要】：目的探討人臍帶間充質(zhì)干細胞(human umbilical cord mesenchymal stem cells,h UC-MSCs)對類噬血細胞性淋巴組織細胞增多癥(hemophagocytic lymphohistocytosis,HLH)小鼠模型肝臟損傷的修復(fù)作用及相關(guān)機制。方法采用h UC-MSCs的體外標(biāo)記,將成年野生型C57BL/6小鼠54只,按完全隨機方法分為模型損傷組(PBS組)、干細胞治療組(MSC組)、正常對照組(CON組)。類HLH小鼠模型采用CPG-ODN1826聯(lián)合IFN-γ誘導(dǎo)產(chǎn)生,MSC組治療采用h UC-MSCs經(jīng)尾靜脈移植。觀察移植后第1、3、7天小鼠一般情況,血生化(ALT、AST、ALB、TG),外周血三系(WBC、RBC、PLT、HGB)變化,第7天肝質(zhì)量體質(zhì)量比,HE染色觀察肝臟組織病理變化,免疫組化法觀察第7天肝臟中活化巨噬細胞的數(shù)量,熒光顯微鏡觀察MSC組CM-Dil標(biāo)記的h UC-MSCs的肝內(nèi)定植,免疫熒光法觀察第7天MSC組肝臟中抗人白蛋白抗體陽性細胞的數(shù)量。結(jié)果與PBS組相比,MSC組小鼠的狀態(tài)好轉(zhuǎn),肝質(zhì)量體質(zhì)量比顯著下降(P0.05);ALT、AST、TG顯著降低(P0.05),ALB無明顯升高(P0.05),WBC、PLT、RBC、HGB顯著升高(P0.05);肝臟炎癥浸潤減少、肝組織損傷恢復(fù);肝臟中活化巨噬細胞數(shù)量明顯減少(P0.05);肝臟中均可發(fā)現(xiàn)有CM-Dil標(biāo)記的h UC-MSCs的定植,且無人白蛋白的表達。結(jié)論 CM-Dil標(biāo)記的h UC-MSCs經(jīng)尾靜脈移植可以在類HLH小鼠受損的肝臟中定植,并可改善肝功能、修復(fù)肝結(jié)構(gòu)、降低活化巨噬細胞的數(shù)量,且不是通過h UC-MSCs肝樣轉(zhuǎn)化實現(xiàn)的。
[Abstract]:Objective to investigate the repair effect of human umbilical cord mesenchymal stem cells (human umbilical cord mesenchymal stem cells) on liver injury in hemophagocytic lymphoid histiocytosis (HLH) mice. Methods 54 adult wild-type C57BL / 6 mice were labeled with hUC-MSCs in vitro and randomly divided into model injury group (PBS group), stem cell therapy group (MSC group) and normal control group (Con group). HLH-like mouse model was treated by tail vein transplantation of hUC-MSCs induced by CPG-ODN1826 and IFN- 緯. The changes of blood biochemistry (alt) and peripheral blood triplex (WBC) were observed on the 7th day after transplantation. The pathological changes of liver tissue were observed by HE staining on the 7th day, and the number of activated macrophages in the liver on the 7th day was observed by immunohistochemical method. The intrahepatic colonization of hUC-MSCs labeled with CM-Dil in MSC group was observed by fluorescence microscope, and the number of anti-albumin antibody positive cells in liver of MSC group was observed by immunofluorescence method on the 7th day. Results compared with PBS group, the condition of MSC group was improved, the liver mass ratio was significantly decreased (P0.05), the ALB level was not significantly increased (P0.05), the liver inflammation infiltration was decreased, liver tissue injury was recovered, and there was no significant increase in ALB (P0.05). The number of activated macrophages in the liver was significantly decreased (P0.05), and CM-Dil labeled hUC-MSCs were found in the liver with no albumin expression. Conclusion CM-Dil labeled hUC-MSCs can be transplanted into the damaged liver of HLH-like mice through tail vein, and can improve liver function, repair liver structure and reduce the number of activated macrophages.
