年齡對(duì)獼猴骨髓間充質(zhì)干細(xì)胞體外分化和細(xì)胞因子水平的影響

【摘要】:背景:間充質(zhì)干細(xì)胞因其良好的生物學(xué)特性和廣泛的應(yīng)用前景而受到關(guān)注,但隨著個(gè)體衰老和體外培養(yǎng)時(shí)間的增加,細(xì)胞逐漸呈現(xiàn)衰老的特征。非人靈長(zhǎng)類動(dòng)物與人具有相似的生物學(xué)特性,在動(dòng)物模型和疾病治療研究方面具有獨(dú)特的優(yōu)勢(shì)。目的:分析不同年齡段獼猴骨髓間充質(zhì)干細(xì)胞的增殖和分化情況,探討年齡對(duì)骨髓間充質(zhì)干細(xì)胞的影響及可能機(jī)制。方法:通過(guò)骨髓穿刺術(shù)采集3歲以內(nèi)和20歲以上雄性獼猴的骨髓,分為幼年組和老年組,每組3只,分離骨髓間充質(zhì)干細(xì)胞進(jìn)行體外培養(yǎng),觀察其形態(tài)變化、增殖能力和分化能力,進(jìn)行衰老相關(guān)的β-半乳糖苷酶染色,ELISA法和蛋白芯片檢測(cè)細(xì)胞因子水平。結(jié)果與結(jié)論:隨著年齡的增長(zhǎng),老年獼猴骨髓間充質(zhì)干細(xì)胞的增殖、分化能力顯著降低,衰老細(xì)胞顯著增加,細(xì)胞因子白細(xì)胞介素1b、白細(xì)胞介素4、白細(xì)胞介素6、腫瘤壞死因子α和血管內(nèi)皮生長(zhǎng)因子水平顯著降低,肝素結(jié)合堿性成纖維細(xì)胞生長(zhǎng)因子和胎盤生長(zhǎng)因子水平顯著升高,說(shuō)明骨髓間充質(zhì)干細(xì)胞的增殖、分化、旁分泌等功能會(huì)因衰老而逐漸受到損害。
[Abstract]:Background: mesenchymal stem cells (MSCs) have attracted much attention due to their good biological characteristics and wide application prospects. However, with the increase of individual senescence and culture time in vitro, the cells gradually present the characteristics of senescence. Nonhuman primates have similar biological characteristics to humans and have unique advantages in animal models and disease therapy. Aim: to analyze the proliferation and differentiation of bone marrow mesenchymal stem cells (BMSCs) in rhesus monkeys at different ages, and to explore the effect of age on bone marrow mesenchymal stem cells (BMSCs) and its possible mechanism. Methods: bone marrow of male rhesus monkeys aged less than 3 years old and over 20 years old were collected by bone marrow puncture and divided into juvenile group and senile group. Bone marrow mesenchymal stem cells (BMSCs) were isolated and cultured in vitro, and their morphological changes were observed. The levels of cytokines were detected by 尾 -galactosidase staining Elisa and protein chip assay. Results and conclusion: the proliferation and differentiation ability of bone marrow mesenchymal stem cells in old rhesus macaques were significantly decreased and the aging cells were significantly increased with the increase of age. The levels of cytokines interleukin 1b, interleukin 4, interleukin 6, tumor necrosis factor 偽 and vascular endothelial growth factor decreased significantly, while heparin binding basic fibroblast growth factor and placental growth factor increased significantly. These results indicate that the proliferation, differentiation and paracrine function of bone marrow mesenchymal stem cells are gradually impaired by aging.
