【摘要】：目的研究中國恒河猴猴免疫缺陷病毒(SIV)mac239急性期感染模型外,周血CD4+T淋巴細胞(簡稱CD4細胞)、淋巴結CD4細胞和血清甘露糖結合凝集素(MBL)的變化情況,探索感染早期固有免疫與適應性免疫的病理變化。方法選用5只健康中國恒河猴,靜脈接種SIVmac239病毒株,分別于感染前、感染后2周、4周和10周檢測血常規(guī)、血漿病毒載量,流式細胞儀分析外周血淋巴細胞亞群,免疫組織化學染色法觀察淋巴結CD4細胞,酶聯(lián)免疫吸附試驗法檢測血清MBL。結果感染后CD4細胞比值較感染前降低27.3%(P0.05),CD8細胞比值較感染前升高44.3%(P0.05),外周血CD4細胞絕對數(shù)下降23.6%,淋巴結CD4細胞上升14.9%,但是差異無統(tǒng)計學意義。血清MBL在感染后10周內持續(xù)下降至感染前水平的24.5%(P0.01)。結論 SIV急性感染期固有免疫破壞顯著,適應性免疫主要表現(xiàn)為體液免疫與細胞免疫的失衡。
[Abstract]:Objective to study the changes of peripheral blood CD4 T lymphocytes (CD4 cells, CD4 cells and serum mannose binding lectin) in Chinese rhesus monkey (Rhesus monkey) in acute stage infection model. To explore the pathological changes of innate immunity and adaptive immunity in the early stage of infection. Methods five healthy Chinese rhesus monkeys were inoculated intravenously with SIVmac239 virus strain. Blood routine examination, plasma viral load and peripheral blood lymphocyte subsets were detected before infection, 2 weeks and 10 weeks after infection, respectively. Lymph node CD4 cells were observed by immunohistochemical staining and serum CD4 was detected by enzyme linked immunosorbent assay (Elisa). Results after infection, the ratio of CD4 cells to CD8 cells was decreased by 27.3% than that before infection. The ratio of P0.05 cells was increased by 44.3%, the absolute number of CD4 cells in peripheral blood decreased by 23.6T, and the CD4 cells of lymph nodes increased by 14.9B, but the difference was not statistically significant. The level of serum MBL decreased to 24.5g / 10 weeks after infection (P 0.01). Conclusion the inherent immune damage in acute infection of SIV is significant, and the imbalance between humoral immunity and cellular immunity is the main manifestation of adaptive immunity.
