滋陰明目丸對(duì)視網(wǎng)膜光損傷大鼠模型Bcl-2及p53表達(dá)的影響

【摘要】:目的:通過觀察滋陰明目丸對(duì)視網(wǎng)膜光損傷大鼠模型Bcl-2、p53表達(dá)的影響,探討滋陰明目丸對(duì)視網(wǎng)膜光損傷大鼠模型的保護(hù)作用及治療機(jī)制。方法:將60只Sprague-Dawley大鼠隨機(jī)分為空白對(duì)照組、模型對(duì)照組、陽(yáng)性對(duì)照組、滋陰明目丸低劑量組、滋陰明目丸中劑量組、滋陰明目丸高劑量組6組,每組10只?瞻讓(duì)照組不予處理,其余均建立視網(wǎng)膜光損傷大鼠模型,造模后第3天予以灌胃,連續(xù)15 d,其中滋陰明目丸低、中、高劑量組予以不同劑量滋陰明目丸灌胃,陽(yáng)性對(duì)照組予以杞菊地黃丸灌胃。取大鼠眼球做免疫組化檢測(cè)Bcl-2及p53的表達(dá)。結(jié)果:Bcl-2免疫組化結(jié)果:與空白對(duì)照組比較,模型對(duì)照組有統(tǒng)計(jì)學(xué)差異(P=0.0250.05);與模型對(duì)照組比較,陽(yáng)性對(duì)照組(P=0.0030.01)、中劑量組(P=0.0040.01)及高劑量組(P=0.0010.01)均有顯著統(tǒng)計(jì)學(xué)差異,低劑量組無(wú)明顯差異(P0.05)。p53免疫組化結(jié)果:與空白對(duì)照組比較,模型對(duì)照組有顯著統(tǒng)計(jì)學(xué)意義(P=0.0000.01);與模型對(duì)照組比較,低劑量組無(wú)明顯差異(P0.05);陽(yáng)性對(duì)照組(P=0.012)、中劑量組(P=0.002)及高劑量組(P=0.002)均有統(tǒng)計(jì)學(xué)差異(P0.05)。結(jié)論:視網(wǎng)膜光損傷模型可使大鼠視網(wǎng)膜組織抗凋亡蛋白Bcl-2表達(dá)降低,促凋亡蛋白p53表達(dá)增高,滋陰明目丸能有效增強(qiáng)Bcl-2的表達(dá),降低p53的表達(dá),從而對(duì)細(xì)胞凋亡起到雙向調(diào)節(jié)作用,對(duì)視網(wǎng)膜光損傷模型具有一定的治療作用。
[Abstract]:Objective: to investigate the protective effect and therapeutic mechanism of Ziyin Mingmu pill (ZYMW) on the expression of Bcl-2 p53 in rat model of retinal light injury. Methods: sixty Sprague-Dawley rats were randomly divided into blank control group, model control group, positive control group, low dose group, middle dose group and high dose group, with 10 rats in each group. No treatment was given in the blank control group. The other rats were given intragastric administration of Ziyin Mingmu pills for 15 days on the 3rd day after the establishment of the model. The middle and high dose groups were given different doses of Ziyin Mingmu pills. The positive control group was given Qiju Dihuang pills by stomach. The expression of Bcl-2 and p53 were detected by immunohistochemistry. Results: compared with the control group, the model control group had statistical difference (P < 0.01), the positive control group (P < 0.0030.01), the middle dose group (P < 0.0040.01) and the high dose group (P < 0.0010.01). There was no significant difference between the low dose group and the model control group (P 0.05N. P53 immunohistochemical results): compared with the blank control group, the model control group had significant statistical significance, and the model control group had significant statistical significance compared with the model control group. There was no significant difference between the low dose group and the high dose group (P 0.05), but there was a significant difference in the positive control group (P 0. 012), the middle dose group (P 0. 002) and the high dose group (P 0. 002). Conclusion: the model of retinal light injury can decrease the expression of anti-apoptotic protein Bcl-2 and increase the expression of apoptotic protein p53 in rat retina. Ziyin Mingmu pills can effectively enhance the expression of Bcl-2 and decrease the expression of p53. Thus, apoptosis plays a bidirectional regulatory role, and has a certain therapeutic effect on retinal light injury model.
