【摘要】：目的:提取小鼠骨髓樹(shù)突狀細(xì)胞(DCs),體外給予丹參酮ⅡA干預(yù),觀察藥物刺激后DCs功能的改變,從而探討丹參酮ⅡA在免疫系統(tǒng)中的作用機(jī)制。方法:提取小鼠的骨髓DCs,體外給予10 ng/ml GM-CSF及IL-4的完全培養(yǎng)液培養(yǎng),并在第5天,磁珠分選得到純度90%以上的樹(shù)突狀細(xì)胞,體外給予一定濃度的丹參酮ⅡA及LPS刺激,收集細(xì)胞及上清,運(yùn)用流式細(xì)胞技術(shù)檢測(cè)DCs表型,ELISA方法檢測(cè)細(xì)胞上清TNF-α、IL-12含量變化,同種混合淋巴細(xì)胞反應(yīng)檢測(cè)樹(shù)突狀細(xì)胞刺激淋巴細(xì)胞增殖及分化的能力。結(jié)果:在丹參酮ⅡA濃度為500 ng/ml時(shí),藥物對(duì)DCs抑制作用達(dá)到最大,因此選取該濃度為實(shí)驗(yàn)作用濃度;即在500 ng/ml作用下,實(shí)驗(yàn)組與對(duì)照組相比,DCs表達(dá)MHCⅡ、CD86及CD80水平均顯著降低(P0.05);實(shí)驗(yàn)組DCs分泌的TNF-α及IL-12含量均顯著降低(P0.05);實(shí)驗(yàn)組DCs刺激淋巴細(xì)胞增殖反應(yīng)能力明顯降低(P0.05);實(shí)驗(yàn)組DCs刺激T淋巴細(xì)胞分泌IL-4含量明顯高于對(duì)照組,IFN-γ含量明顯低于對(duì)照組(P0.05)。結(jié)論:丹參酮ⅡA可以通過(guò)降低LPS誘導(dǎo)的DCs成熟狀態(tài),來(lái)參與免疫系統(tǒng)或自身免疫性疾病的發(fā)生發(fā)展。
[Abstract]:Objective: to extract murine bone marrow derived dendritic cells (DCs) in vitro, given tanshinone A intervention, change DCs function observed after drug stimulation, so as to explore the mechanism of tanshinone II A in immune system. Methods: the extraction of mouse bone marrow DCs, completely cultured in vitro treated with 10 ng/ml GM-CSF and IL-4 in liquid culture, and in fifth days, by magnetic sorting of dendritic cells more than 90% purity, tanshinone A and LPS in vitro with certain concentration of stimulation, collect the cells and supernatant were detected by flow cytometry. DCs phenotype, ELISA method to detect the cell supernatant of TNF- alpha, IL-12 content, mixed lymphocyte reaction and detection ability to stimulate the proliferation of dendritic cells the differentiation of lymphocytes. Results: tanshinone II A concentration was 500 ng/ml, the drug inhibition of DCs reached the maximum, so choose the concentration of experimental concentration; in 500 ng/ml, Compared to the experimental group and the control group, the expression of DCs MHC II, CD86 and CD80 levels were significantly decreased (P0.05); TNF- alpha and IL-12 content in the experimental group significantly decreased the secretion of DCs (P0.05); experimental group DCs stimulated lymphocyte proliferation were significantly decreased (P0.05); experimental group DCs stimulated T secretion of IL-4 content significantly higher than the control group, IFN- gamma content was significantly lower than the control group (P0.05). Conclusion: tanshinone A can reduce LPS induced DCs maturation, the immune system or to participate in the occurrence and development of autoimmune diseases.
