【摘要】：目的制備貓視神經(jīng)的連續(xù)切片,選擇合適的染色方法 ,尋找合適的配準(zhǔn)方法 ,經(jīng)圖像采集及初步處理后利用計(jì)算機(jī)的三維重建技術(shù),獲得可視化的貓視神經(jīng)三維結(jié)構(gòu)。方法利用石蠟切片技術(shù)的制備貓視神經(jīng)連續(xù)切片,采用特殊的神經(jīng)三色染色法進(jìn)行切片染色。利用數(shù)碼顯微鏡及Motic Images Advanced 3.0和Motic Images Assembly 1.0軟件對(duì)連續(xù)的貓視神經(jīng)石蠟切片進(jìn)行拍照,獲得貓視神經(jīng)連續(xù)切片的原始圖像,將原始圖像在GIMP軟件中經(jīng)過(guò)調(diào)整亮度對(duì)比度、初步圖像配準(zhǔn)、提取神經(jīng)束膜圖像信息后在Amira軟件下,經(jīng)過(guò)計(jì)算機(jī)在手工配準(zhǔn)的基礎(chǔ)上進(jìn)一步自動(dòng)配準(zhǔn),把二維的切片圖像重建成三維的可視化圖像。結(jié)果實(shí)驗(yàn)所得經(jīng)三色染色的連續(xù)切片的原始圖像上神經(jīng)束膜能較好的與周?chē)慕M織區(qū)分;利用Amira三維重建軟件,重建得到貓視神經(jīng)三維結(jié)構(gòu)圖像,重建后的貓視神經(jīng)結(jié)構(gòu)三維圖像可進(jìn)行任意的切割、旋轉(zhuǎn)、回復(fù)、拍照等操作。結(jié)論實(shí)驗(yàn)證明在不利用標(biāo)志線(xiàn)的條件下,通過(guò)制備視神經(jīng)連續(xù)切片,經(jīng)過(guò)染色拍照,手工圖像配準(zhǔn)后利用計(jì)算機(jī)自動(dòng)配準(zhǔn)技術(shù)對(duì)視神經(jīng)進(jìn)行三維重建是完全可行的。
[Abstract]:Objective to prepare the serial sections of the optic nerve of the cat, to select the appropriate staining method and to find the appropriate registration method, and to obtain the visual three-dimensional structure of the optic nerve of the cat by using the three-dimensional reconstruction technique of the computer after the image acquisition and preliminary processing.Methods Cat optic nerve serial sections were prepared by paraffin technique and stained by special nerve trichrome staining.After the initial image registration and the extraction of the neural bundle membrane image information, the two-dimensional slice image is reconstructed into a three-dimensional visual image by computer automatic registration on the basis of manual registration under the Amira software.Results the nerve trachea on the original images of the serial slices stained by three colors could be distinguished from the surrounding tissues, the three-dimensional reconstruction software of Amira was used to reconstruct the three-dimensional structure images of the optic nerve of the cat.The reconstructed 3D image of optic nerve structure of cat can be cut, rotated, restored, photographed and so on.Conclusion it is proved by experiments that it is feasible to reconstruct the optic nerve by computer automatic registration technique by making continuous sections of the optic nerve without using the mark line and taking pictures by staining and taking pictures by hand.
