【摘要】：目的探討結(jié)核分枝桿菌感染對(duì)巨噬細(xì)胞內(nèi)膽固醇酯的影響。方法將小鼠RAW264.7細(xì)胞分為空白對(duì)照組和結(jié)核分枝桿菌感染(細(xì)菌數(shù)與細(xì)胞數(shù)之比分別為5∶1、10∶1、20∶1)實(shí)驗(yàn)組,孵育1、3、5、7d后采用油紅O染色觀察細(xì)胞內(nèi)脂滴的變化,利用酶法測(cè)定細(xì)胞內(nèi)總膽固醇、游離膽固醇和膽固醇酯相對(duì)含量的變化。結(jié)果細(xì)菌數(shù)與細(xì)胞數(shù)之比為20∶1孵育1d,油紅O染色細(xì)胞內(nèi)可見(jiàn)大量脂滴,膽固醇酯相對(duì)含量為(65.91±2.45)%,孵育7d,大量細(xì)胞出現(xiàn)裂解,脂滴溢出。細(xì)胞內(nèi)膽固醇酯的含量隨時(shí)間和菌量的增加而升高。結(jié)論結(jié)核分枝桿菌感染可以導(dǎo)致小鼠巨噬細(xì)胞內(nèi)膽固醇酯含量增加,且與時(shí)間和菌量成正比。
[Abstract]:Objective to investigate the effect of Mycobacterium tuberculosis infection on cholesterol ester in macrophages. Methods RAW264.7 cells in mice were divided into blank control group and Mycobacterium tuberculosis infection group (the ratio of bacterial number to cell number was 5: 1 10: 1: 20: 1). Oil red O staining was used to observe the changes of lipid droplets and total cholesterol in the cells was determined by enzymatic method after incubation for 7 days. Results when the ratio of bacterial number to cell number was 1 day at 20:1, a large number of lipid droplets could be found in the cells stained with oil red O, and the relative content of cholesterol ester was 65.91 鹵2.45%. After incubation for 7 days, a large number of cells were lysed. Lipid spill. The content of cholesterol ester in cells increased with the increase of time and bacteria. Conclusion Mycobacterium tuberculosis infection can lead to the increase of cholesterol ester content in mouse macrophages, which is directly proportional to the time and the amount of bacteria.
