結(jié)核分枝桿菌Rv3245蛋白抗原免疫學(xué)特性的研究

【摘要】:目的:研究結(jié)核分枝桿菌Rv3425蛋白免疫學(xué)特性,評估其診斷應(yīng)用價值及在結(jié)核致病性中的作用。方法:將Rv3425基因克隆至p ET28a載體中,誘導(dǎo)表達Rv3425融合蛋白并進行純化;利用ELISA、Western blot分析其抗原性與特異性;流式檢測該抗原對巨噬細胞凋亡的影響。結(jié)果:獲得了可溶性原核表達融合蛋白Rv3425,Rv3425蛋白能刺激滅活結(jié)核分枝桿菌免疫小鼠脾細胞產(chǎn)生高水平的特異性IFN-γ、純化的Rv3425蛋白能與結(jié)核感染小鼠血清特異性結(jié)合,其特異性血清抗體Ig G及Ig M在結(jié)核病人中的水平明顯高于健康人,發(fā)現(xiàn)該蛋白能誘導(dǎo)巨噬細胞的凋亡。結(jié)論:本研究發(fā)現(xiàn)結(jié)核分枝桿菌Rv3425蛋白具有較強的抗原性且能促進巨噬細胞的凋亡,這些發(fā)現(xiàn)對于結(jié)核病的診斷及致病機制的研究具有重要價值。
[Abstract]:Objective: to study the immunological characteristics of Rv3425 protein of Mycobacterium tuberculosis and to evaluate its diagnostic value and its role in the pathogenicity of tuberculosis. Methods: the Rv3425 gene was cloned into p ET28a vector to express and purify the Rv3425 fusion protein. The antigenicity and specificity of EISAL were analyzed by Western blot. Results: the soluble prokaryotic expression fusion protein Rv3425rv3425 could stimulate the production of high specific IFN- 緯 by inactivated Mycobacterium tuberculosis immunized mice spleen cells and purified Rv3425. The protein can specifically bind to the serum of mice infected with tuberculosis. The levels of IgG and IgM in tuberculosis patients were significantly higher than those in healthy persons. Conclusion: the Rv3425 protein of Mycobacterium tuberculosis has strong antigenicity and can promote the apoptosis of macrophages. These findings are of great value in the diagnosis and pathogenesis of tuberculosis.
