【摘要】：目的:建立小鼠流感性肺炎模型,明確呼吸道流感病毒感染過(guò)程中是否存在特異性的腸道病理?yè)p傷,并探討了其在"肺與大腸相表里"研究中的應(yīng)用。方法:BALB/c小鼠48只隨機(jī)分為正常組、病毒組。病毒組給予50μL、50LD50病毒液滴鼻建立流感病毒感染的小鼠肺炎模型,感染后第3、5、6天分別取材,HE染色檢測(cè)肺臟、大腸、小腸、心臟、肝臟、腎臟組織病理結(jié)果,血生化檢測(cè)血清谷草轉(zhuǎn)氨酶(AST)、谷丙轉(zhuǎn)氨酶(ALT)和尿素氮(BUN)水平。結(jié)果:流感病毒感染后,病毒組小鼠肺組織出現(xiàn)明顯的炎性病變外,隨時(shí)間延長(zhǎng)肺組織充血水腫出血加重;小腸黏膜腺體排列紊亂,絨毛粗大、部分脫落,黏膜固有層炎性細(xì)胞浸潤(rùn);大腸黏膜固有層、肌層大量炎性細(xì)胞浸潤(rùn),腺體結(jié)構(gòu)消失、脫落;而心臟、肝臟、腎臟無(wú)明顯病理改變;病毒組小鼠血清AST、ALT、BUN較正常組相比,差異無(wú)統(tǒng)計(jì)學(xué)意義。結(jié)論:小鼠流感病毒性肺炎模型中存在小腸、大腸特異性免疫病理?yè)p傷,是研究中醫(yī)"肺與大腸相表里"理論及相關(guān)臨床治法的理想模型。
[Abstract]:Objective: to establish a mouse model of influenza pneumonia and determine whether there is specific intestinal pathological injury in the process of respiratory influenza virus infection. In this paper, we also discussed its application in the study of "lung and large intestine". Methods Forty-eight BALB / c mice were randomly divided into normal group and virus group. The viral group was given 50 渭 L of 50LD50 virus solution to establish the pneumonia model of influenza virus infection in mice. On the 3rd 5th day after infection, HE staining was used to detect the pathological results of lung, large intestine, small intestine, heart, liver and kidney, and the serum levels of aspartate, alanine aminotransferase (alt) and urea nitrogen bun (bun) were detected in blood biochemistry. Results: after infection of influenza virus, serum alanine aminotransferase (alt) and urea nitrogen bun (bun) were detected. In the viral group, there were obvious inflammatory lesions in the lung tissue, and the hyperemia and edema and hemorrhage of the lung tissue became more severe with the prolongation of time, the intestinal mucosal glands arranged in disorder, the villi were coarse, some of them fell off, the inflammatory cells of the lamina propria were infiltrated, and the lamina propria of the large intestine, A large number of inflammatory cells infiltrated in the myometrium, the structure of the glands disappeared and shed off, while the heart, liver and kidney had no obvious pathological changes. The serum ASTT alt bun in the virus group was higher than that in the normal group. Conclusion: there are small intestine and large intestine specific immunopathologic damage in mouse model of influenza virus pneumonia, which is an ideal model for studying the theory of "lung and large intestine" and related clinical treatment methods in traditional Chinese medicine.
