微小巴貝斯蟲烯醇化酶蛋白主要特征與抗原表位的生物信息學分析

【摘要】:目的運用生物信息學方法分析微小巴貝斯蟲烯醇化酶(BmEno)基因編碼蛋白的主要特征和抗原表位。方法運用ORF Finder、PROSITE、ProtPar am、SignalP、TMpred、SOSUI、MotifScan、Bcepred、HNN等生物信息學在線分析軟件,結(jié)合DNASTAR生物信息學分析軟件,預測BmEno蛋白的開放閱讀框、特征結(jié)構(gòu)域、理化性質(zhì)、信號肽、跨膜區(qū)、翻譯后修飾位點、可溶性、親水性、表面可及性、二級結(jié)構(gòu)和抗原表位。結(jié)果該蛋白由438個氨基酸組成,在349-362位存在烯醇化酶的特征結(jié)構(gòu)域,分子式為C_(2098)H_(3366)N_(570)O_(648)S_(18),分子質(zhì)量單位為47.5203ku,等電點理論值為5.98,有25個翻譯后修飾位點,10個親水性參數(shù)得分≥1.9的區(qū)域,6個柔韌性參數(shù)得分≥2.0的區(qū)域,18個表面可及性參數(shù)得分≥1.9的區(qū)域,7個潛在的B細胞抗原表位,15個潛在的T細胞抗原表位,為可溶性蛋白。結(jié)論微小巴貝斯蟲烯醇化酶蛋白為可溶性蛋白,有多個抗原表位,具有潛在的免疫原性,可以作為微小巴貝斯蟲的疫苗候選抗原。
[Abstract]:Objective to analyze the main characteristics and antigenic epitopes of BmEnogene encoding protein by bioinformatics. Methods ORF Findern proside was used. The bioinformatics online analysis software, such as ProtPar signal Pnn TMpredl, motif Scann BcepredHNN and so on, is used to analyze bioinformatics on line. Combined with DNASTAR bioinformatics software, the open reading frame, characteristic domain, physicochemical properties, signal peptide, transmembrane region, post-translational modification site, soluble and hydrophilic of BmEno protein were predicted. Results the protein was composed of 438 amino acids and had the characteristic domain of enolase at 349-362. The molecular formula is C / T / 2098H / C / C / C / C / T / T / T / T / T / T / C / C / C / C / C / C / C / C / C / C / C / C / C / C / C / C / C / C / C / C / C / C / C / C / C / C / C / C / C / C.
