豬卵母細(xì)胞成熟和早期胚胎發(fā)育過(guò)程中的microRNA表達(dá)譜研究

【摘要】:豬是廣泛飼養(yǎng)的經(jīng)濟(jì)動(dòng)物,提供重要的肉食品來(lái)源;也是重要的實(shí)驗(yàn)動(dòng)物,是人類(lèi)醫(yī)學(xué)的試驗(yàn)?zāi)P椭弧icroRNAs (miRNAs)參與如細(xì)胞增殖、分化等許多生物學(xué)過(guò)程的調(diào)控。已有的研究發(fā)現(xiàn),在許多物種的卵母細(xì)胞成熟和早期胚胎發(fā)育過(guò)程中,miRNA的表達(dá)豐度都是呈現(xiàn)動(dòng)態(tài)變化的。miRNA可影響卵母細(xì)胞成熟、早期胚胎卵裂、母源mRNA降解等過(guò)程。但是,目前對(duì)于豬卵母細(xì)胞成熟和早期胚胎發(fā)育過(guò)程中miRNA的研究還相對(duì)較少。本研究選取GV期卵母細(xì)胞、MII期卵母細(xì)胞、4-細(xì)胞胚胎和囊胚四個(gè)時(shí)間點(diǎn)的樣本,采用solexa測(cè)序的方法對(duì)miRNA的表達(dá)譜進(jìn)行系統(tǒng)分析,篩選出了在豬卵母細(xì)胞成熟及早期胚胎發(fā)育過(guò)程中有重要作用的miRNAs,在對(duì)差異表達(dá)的miRNAs的靶基因進(jìn)行預(yù)測(cè)后,還對(duì)這些預(yù)測(cè)的靶基因進(jìn)行通路分析。此外,通過(guò)Q-PCR驗(yàn)證solexa測(cè)序結(jié)果的同時(shí),對(duì)關(guān)鍵]miRNA如mir-27、let-7i等進(jìn)行進(jìn)一步試驗(yàn)研究,為闡明豬卵母細(xì)胞成熟及早期胚胎發(fā)育過(guò)程中miRNA的調(diào)控機(jī)制奠定了基礎(chǔ)。
[Abstract]:Pigs are widely raised economic animals and provide important sources of meat food. They are also important experimental animals. MicroRNAs (miRNAs) are involved in many biological processes such as cell proliferation and differentiation. It has been found that the abundance of miRNA expression in oocyte maturation and early embryonic development of many species is a dynamic change. MiRNA may affect oocyte maturation, early embryo cleavage, and the degradation of mother mRNA. However, there are few studies on miRNA in pig oocytes maturation and early embryonic development. In this study, the expression profiles of miRNAs were systematically analyzed by solexa sequencing from four time points of GV phase oocyte phase II oocytes, embryos and blastocysts. MiRNAss, which play an important role in the maturation and early embryonic development of porcine oocytes, were screened. After predicting the target genes of differentially expressed miRNAs, the pathway of these predicted genes was analyzed. In addition, the solexa sequencing results were verified by Q-PCR, and the key miRNAs, such as mir-27let-7i, were further studied, which laid a foundation for elucidating the regulation mechanism of miRNAs during maturation and early embryonic development of porcine oocytes.
