【摘要】:癌癥是威脅人類健康的一大殺手,其中宮頸癌是威脅女性的第二大惡性疾病。研究數(shù)據(jù)表明,對(duì)于早期癌癥的治療效果明顯優(yōu)于中晚期癌癥。這就意味著如果能在癌癥初期對(duì)其進(jìn)行準(zhǔn)確的診斷,就能改善病人的生存質(zhì)量甚至可以延長(zhǎng)病人的生存年限。對(duì)于癌癥的早期診斷,目前主要的方法是基于免疫組織化學(xué)、分子生物學(xué)、流式細(xì)胞術(shù)等檢測(cè)腫瘤標(biāo)記物的病理學(xué)檢測(cè)方法。但這些方法都存在著假陽(yáng)性和假陰性的問(wèn)題。原子力顯微鏡技術(shù)由于具有納米及微納米級(jí)分辨率以及對(duì)細(xì)胞無(wú)任何損傷的特點(diǎn)而得到了廣泛的應(yīng)用。本論文通過(guò)這項(xiàng)技術(shù),檢測(cè)宮頸癌細(xì)胞的性狀變化,進(jìn)而分析出細(xì)胞內(nèi)部的病理變化,達(dá)到癌細(xì)胞早期診斷的效果。同時(shí),為了探索宮頸癌的治療辦法,本文進(jìn)一步研究了經(jīng)抗癌藥多西他賽處理后的宮頸癌細(xì)胞的性狀變化。本文利用原子力顯微鏡在相同的測(cè)量條件下分別對(duì)藥物處理前后的正常宮頸上皮細(xì)胞和宮頸癌HeLa細(xì)胞進(jìn)行了掃描,并對(duì)兩者的力曲線進(jìn)行分析與比較,研究了細(xì)胞的物理化學(xué)性質(zhì)變化。通過(guò)分析得知細(xì)胞經(jīng)過(guò)藥物處理后細(xì)胞的物理化學(xué)性質(zhì)發(fā)生了改變,細(xì)胞的塑性指數(shù)發(fā)生變化,說(shuō)明細(xì)胞的內(nèi)部結(jié)構(gòu)有所改變,這一改變通過(guò)利用Western Blot檢測(cè)細(xì)胞微管蛋白的含量變化得到證明;細(xì)胞的粘滯功發(fā)生改變,細(xì)胞表面的彈性模量發(fā)生變化,這一改變通過(guò)透射電鏡的觀察得到證明,通過(guò)透射電鏡的觀察我們知道細(xì)胞表面的微絨毛經(jīng)過(guò)藥物處理后顯著減少;細(xì)胞的彈性等物理化學(xué)性質(zhì)也投相應(yīng)的改變。通過(guò)AFM測(cè)得的實(shí)驗(yàn)結(jié)果都通過(guò)相關(guān)生物學(xué)實(shí)驗(yàn)進(jìn)一步驗(yàn)證了原子力顯微鏡對(duì)癌細(xì)胞診斷結(jié)果的正確性。原子力顯微鏡對(duì)細(xì)胞的這種無(wú)標(biāo)記檢測(cè)手段彌補(bǔ)了這一領(lǐng)域的空白,使得它在未來(lái)的科學(xué)研究中能起到至關(guān)重要的作用。此外,原子力顯微鏡也有望成為醫(yī)學(xué)檢測(cè)方向的重要檢測(cè)工具。
[Abstract]:Cancer is a major threat to human health, including cervical cancer is the second malignant diseases threatening women. Research data show that in late stage of early cancer treatment is superior in cancer. This means that if you can make accurate diagnosis of the quality of life in the early stage of cancer, can improve the patients can even be extended the patient's survival. For the early diagnosis of cancer, at present, the main method is based on immunohistochemistry, molecular biology, pathology detection method of flow cytometry detection of tumor markers. But these methods exist in the problem of false positives and false negatives. Atomic force microscopy has been widely used because of the characteristics of nano and micro nano resolution of cells without any damage. This paper through this technology, detection of changes of characteristics of cervical cancer cells, and then analyze the fine The pathological changes inside the cell, to achieve the effect of cancer cells in early diagnosis of cervical cancer. At the same time, in order to explore the way of treatment, this paper further studies the changes of characteristics of anticancer drugs docetaxel after treatment of cervical cancer cells. By using atomic force microscopy in the measurement under the same circumstances respectively before and after drug treatment of normal cervical epithelial cells and HeLa cervical cancer cells were scanned, and the stress curves are analyzed and compared, studied the change of physical and chemical properties of cells. Cells that treated cells after the physical and chemical properties changed through analysis, cell plasticity index changes, shows the internal structure of cells changed, the a change has been proved by changing the content of cells was detected by Western Blot tubulin; viscous power cells changed, the elastic modulus of the cell surface occurs Change, this change was demonstrated by transmission electron microscopy, transmission electron microscope and we know that the microvilli were significantly reduced after treated by medicine; physical and chemical properties of the elastic cell also cast the corresponding change. Measured by AFM experimental results through biological experiments further verified by atomic force microscopy the diagnosis results of cancer cells. The atomic force microscope for cell label free detection means to make up for gaps in this field, so that it can play a crucial role in scientific research in the future. In addition, an important tool for detection of atomic force microscope is expected to become the direction of medical testing.
