【摘要】:目的:分析KRT1、KRT10、GSTP1基因在各腫瘤中表達的大數(shù)據(jù)信息及在哈薩克族食管鱗癌組織中的表達及臨床意義。方法:通過www.proteinatlas.org生物學信息公眾平臺,查詢KRT1、KRT10、GSTP1基因在各正常人體組織及各腫瘤中的表達情況;運用Western blot技術(shù)對28對新鮮食管癌組織及相應的癌旁正常組織中的KRT1、KRT10、GSTP1表達情況進行半定量分析;用免疫組織化學技術(shù)(IHC)對60對食管鱗狀細胞癌組織及癌旁正常食管黏膜組織中的KRT1、KRT10、GSTP1表達情況進行定性檢測,結(jié)合臨床病理資料,對免疫組化結(jié)果進行數(shù)據(jù)分析,并用ROC曲線對三個蛋白的診斷價值進行評估。結(jié)果:1.Western blot結(jié)果顯示KRT1、KRT10、GSTP1三個蛋白在食管鱗狀細胞癌組織中表達高于相應癌旁正常組織(P0.001),在哈薩克族與漢族中表達無統(tǒng)計學差異(P0.05);2.免疫組化結(jié)果顯示KRT1、KRT10、GSTP1在食管鱗狀細胞癌組織中的表達均高于癌旁正常組織(P0.001),結(jié)合臨床病理參數(shù)分析,結(jié)果顯示KRT1、KRT10的表達與食管鱗癌組織的分化程度有關(guān)(P0.05),GSTP1在食管鱗狀細胞癌中的強陽性表達與患者的年齡、性別、族別、淋巴結(jié)轉(zhuǎn)移與否有關(guān)(P0.05),根據(jù)ROC曲線下面積提示三個差異蛋白中KRT10蛋白的診斷價值最高,GSTP1蛋白其次,KRT1蛋白最低,三者均有診斷價值。結(jié)論:KRT1、KRT10、GSTP1為食管鱗狀細胞癌的發(fā)生發(fā)展中的重要變化分子,KRT1、KRT10與食管鱗癌的分化有關(guān),可作為食管鱗癌惡性程度判斷的重要指標,KRT10有望成為食管鱗癌診斷中的特異性分子標記物,GSTP1與患者的年齡、性別、族別、淋巴結(jié)轉(zhuǎn)移有關(guān),可對食管鱗癌的協(xié)助診斷、惡性程度判斷及后期個體化治療有一定指導意義。
[Abstract]:Objective: to analyze the large data information expressed by KRT1, KRT10 and GSTP1 genes in various tumors and the expression and clinical significance in Kazak's esophageal squamous cell carcinoma. Methods: the www.proteinatlas.org biological information public platform, the query expression of KRT1, KRT10, GSTP1 gene in normal human tissues and tumor; using semi quantitative analysis of 28 pairs of cancer and corresponding fresh esophageal cancer tissues, the expression of GSTP1 Western KRT1, KRT10 blot; immunohistochemistry Chemical Technology (IHC) expression were detected in 60 of esophageal squamous cell carcinoma tissue and normal esophageal mucosa tissues of KRT1, KRT10, GSTP1, combined with clinical data, data analysis on the results of immunohistochemistry, and were evaluated by ROC curve in diagnosis of three proteins. Results: 1.Western blot showed that KRT1, KRT10 and GSTP1 three protein expression in esophageal squamous cell carcinoma tissues than adjacent normal tissues (P0.001), there were no significant difference in Kazak and Han in 2. (P0.05); immunohistochemistry showed that the expression of KRT1, KRT10 and GSTP1 in esophageal squamous cell carcinoma were higher than those in adjacent normal tissues (P0.001), combined with analysis of clinical pathological parameters, the results showed that KRT1, KRT10 expression and differentiation of esophageal squamous cell carcinoma (P0.05), GSTP1 strongly positive in esophageal squamous cell carcinoma and patients' age, gender, nationality, is associated with lymph node metastasis (P0.05), according to the diagnostic value of the area under the ROC curve showed that KRT10 protein three proteins in the highest GSTP1 protein KRT1 protein, secondly, the lowest value of three has diagnosis. Conclusion: KRT1, KRT10, GSTP1 as an important change in the development of molecular differentiation of esophageal squamous cell carcinoma, KRT1, KRT10 and esophageal squamous cell carcinoma, as an important index to judge the malignant degree of esophageal squamous cell carcinoma, KRT10 is expected to become the specific molecular markers for diagnosis of esophageal squamous cell carcinoma, GSTP1 and the age and sex of the patients, nationality and lymph node metastasis of esophageal squamous cell carcinoma, can assist diagnosis and individualized treatment of judging malignant degree and later have a certain significance.
