應用昆蟲分子系統(tǒng)學對法醫(yī)嗜尸性蠅類種屬鑒定的研究

【摘要】:目的 本課題旨在探索可用于法醫(yī)嗜尸性蠅類種屬鑒定的昆蟲分子系統(tǒng)學方法。方法 選擇線粒體DNA(mtDNA)Cytb和ND1基因,PCR擴增,聚丙烯酰胺凝膠電泳銀染顯色分型,用DNA遺傳分析儀對分型產(chǎn)物測序;利用系統(tǒng)進化學軟件和統(tǒng)計學方法對序列進行分析,建立對法醫(yī)嗜尸性蠅類進行種屬鑒定的方法。結(jié)果 對中國3個地區(qū)6種嗜尸性蠅類的mtDNA Cytb和ND1基因進行了擴增、電泳檢測和序列分析;利用統(tǒng)計學的方法,通過系統(tǒng)進化學的分析,建立了這些嗜尸性蠅類的系統(tǒng)進化關(guān)系;結(jié)合系統(tǒng)進化學的分析結(jié)果,初步建立了法醫(yī)嗜尸性蠅類種屬鑒定的方法。結(jié)論 基于mtDNA Cytb和ND1基因,初步建立了法醫(yī)嗜尸性蠅類種屬鑒定的昆蟲分子系統(tǒng)學方法。
[Abstract]:Objective to explore a molecular phylogenetic method for identification of necrophilic flies in forensic medicine. Methods Mitochondrial DNA (mtDNA) Cytb and ND1 gene were amplified by PCR, polyacrylamide gel electrophoresis (page) silver staining was used to type the genotypes, DNA genetic analyzer was used to sequence the typing products, and the sequence was analyzed by systematic chemical software and statistical method. A method for species identification of necrophilic flies in forensic medicine was established. Results the mtDNA Cytb and ND1 genes of 6 species of necrophilic flies in three regions of China were amplified, electrophoretic and sequenced. The phylogenetic relationship of these necrophagous flies was established, and the method of identifying the species of coriophilic flies by forensic medicine was preliminarily established in combination with the results of phylochemical analysis. Conclusion based on mtDNA Cytb and ND1 genes, a molecular phylogenetic method for identification of species of necrophilic flies was established.
