Caspase-7在大鼠顱腦損傷后表達的免疫組織化學研究

【摘要】: 目的:創(chuàng)傷性顱腦損傷是法醫(yī)工作中經常見到的一種損傷,腦損傷后引起的神經細胞死亡通過壞死和凋亡兩種途徑實現。凋亡發(fā)生是一個復雜的,由caspase家族成員介導的蛋白酶級聯反應過程。Caspase-7是caspase級聯反應中的下游分子之一,為由303個氨基酸組成的蛋白質,分子量為35kD,存在于多種細胞系,彌散分布在胞漿內和近膜結構上,是凋亡的最終效應分子之一。正常情況下Caspase-7以酶原的形式存在,在凋亡過程中被其上游分子活化裂解為兩個活性亞單位,作用于底物,使凋亡得以執(zhí)行。早期認為Caspase-7在中樞神經系統(tǒng)凋亡中并無重要作用,但近年的研究提示其似乎在神經細胞的凋亡中扮演重要的角色,故其日益受到關注。本文采用自由落體撞擊模型和SABC法研究大鼠顱腦損傷后caspase-7的IHC表達情況,旨在發(fā)現其表達是否具有規(guī)律性,希望能為法醫(yī)學顱腦損傷的損傷時間推斷補充新的指標。 方法:采用完全隨機分組方法,根據造創(chuàng)后處死時間不同將實驗大鼠隨機分為傷后1h、3h、6h、12h、1d、3d、5d、7d、10d、14d組,另設正常對照組及假手術組。在預定時間分別處死大鼠,取損傷處及其周圍區(qū)域(對照組、假手術組取與實驗組相同的部位),進行HE染色和caspase-7免疫組化SABC法染色,并對染色結果進行光鏡觀察、圖像分析及統(tǒng)計學處理,以了解caspase-7在大鼠顱腦損傷后的表達情況和規(guī)律。
[Abstract]:Objective: traumatic craniocerebral injury is a kind of injury that is often seen in forensic medicine. The death of nerve cells caused by brain injury is realized by two ways: necrosis and apoptosis. The protease cascade process mediated by members of the caspase family. Caspase-7 is one of the downstream molecules in the caspase cascade reaction. It is a 303-amino acid protein with a molecular weight of 35kD.It exists in a variety of cell lines and distributes in the cytoplasm and near membrane structure. Caspase-7 is one of the final effector molecules of apoptosis. Normally, Caspase-7 exists in the form of prozyme and is activated into two active subunits by upstream molecules during apoptosis, acting on the substrate. It is believed that Caspase-7 does not play an important role in central nervous system apoptosis, but recent studies suggest that Caspase-7 may play an important role in neuronal apoptosis. Therefore, it has attracted more and more attention. In this paper, the expression of caspase-7 IHC in rats after craniocerebral injury was studied by free fall impact model and SABC method, in order to find out whether the expression of caspase-7 was regular or not. It is hoped that it can be used to infer the injury time of forensic craniocerebral injury. Methods: the experimental rats were randomly divided into two groups according to the time of death after injury: the rats were randomly divided into three groups: 1 hour after injury, 3 hours after injury, 1 hour after injury, 3 days after injury, 3 days after injury, 5 days after injury, 7 days after 7 days and 10 days after 14 days, and then set up normal control group and sham operation group, and the rats were killed at the scheduled time, and the rats were divided into two groups: the control group and the sham operation group. The site of injury and its surrounding area (control group, sham-operation group and experimental group) were taken for HE staining and caspase-7 immunohistochemical SABC staining. The staining results were observed by light microscopy, image analysis and statistical analysis. To understand the expression and regularity of caspase-7 after craniocerebral injury in rats.
