大鼠心臟鈍力性損傷的法醫(yī)學(xué)實(shí)驗(yàn)研究

【摘要】:目的 建立大鼠心臟鈍力性損傷模型;觀(guān)察損傷后正常區(qū),損傷區(qū)心肌的病理變化;MMP-1和MMP-2的表達(dá)強(qiáng)度出現(xiàn)的規(guī)律;試圖為心臟鈍力性損傷的法醫(yī)學(xué)鑒定提供客觀(guān)的形態(tài)學(xué)依據(jù),并探討其可能的發(fā)生機(jī)制。 方法 健康成年Wistar大鼠50只隨機(jī)分為七組即0h,6h,12h,24h,3天,5天,7天,0h為對(duì)照組:開(kāi)胸,暴露心臟,逐層關(guān)胸,,待各組成活至相應(yīng)時(shí)間時(shí)處死,取材。其余各組為:開(kāi)胸,暴露心臟,打擊,逐層關(guān)胸,到實(shí)驗(yàn)所需時(shí)間取心臟制作標(biāo)本。HE染色,光鏡觀(guān)察;MMP-1及MMP-2免疫組織化學(xué)染色,計(jì)算機(jī)圖象分析,統(tǒng)計(jì)學(xué)分析。 結(jié)果 (1) 隨時(shí)間的延長(zhǎng),MMP-1和MMP-2的表達(dá)強(qiáng)度呈現(xiàn)規(guī)律變化:MMP-1在傷后12h開(kāi)始明顯表達(dá),逐漸增強(qiáng),3d達(dá)高峰;MMP-2在傷后24h開(kāi)始明顯表達(dá),五天達(dá)高峰。(2)HE染色可見(jiàn)心肌組織淤血,出血,炎細(xì)胞浸潤(rùn),時(shí)間長(zhǎng)的可有纖維化。(3)各損傷組圖象分析結(jié)果與對(duì)照組結(jié)果經(jīng)統(tǒng)計(jì)分析p<0.05。 結(jié)論 (1)心臟損傷模型建立較成功;(2)對(duì)照組與實(shí)驗(yàn)組HE染色結(jié)果與免疫組織化學(xué)結(jié)果有較好的一致性,各指標(biāo)的分布規(guī)律說(shuō)明MMP-1及MMP-2可作為檢測(cè)心肌損傷的指標(biāo)加以研究。
[Abstract]:Objective to establish a rat model of cardiac blunt force injury, observe the pathological changes of myocardium in normal area and injured area after injury, and provide objective morphological basis for forensic identification of cardiac blunt force injury. The possible mechanism of its occurrence was also discussed. Methods Fifty healthy adult Wistar rats were randomly divided into seven groups: 0 h, 6 h, 12 h, 24 h, 3 days, 5 days, 7 days and 0 h as the control group: open chest, expose heart, close chest layer by layer, then kill each group after surviving to the corresponding time, the other groups were open chest and exposed heart. Beating, closing the chest layer by layer, taking the time to make heart specimen. HE staining, light microscope observation of MMP-1 and MMP-2 immunohistochemical staining, computer image analysis, statistical analysis. Results (1) the expression intensity of MMP-1 and MMP-2 showed a regular change with the extension of time, and the expression of MMP-1 began to appear at 12 hours after injury, gradually enhanced the expression of MMP-2 at the peak of 3 days after injury, and reached the peak at 24 hours after injury, and on the fifth day, congestion and bleeding of myocardial tissue were observed by HE staining. The results of image analysis of each injury group were compared with those of the control group (p < 0.05). Conclusion 1) the results of HE staining in the control group and the experimental group are in good agreement with the immunohistochemical results. The distribution of each index indicates that MMP-1 and MMP-2 can be used as indicators for the detection of myocardial injury.
