肝缺血再灌注初期控制性降壓對(duì)大鼠腦內(nèi)PPAR-γmRNA表達(dá)的影響

【摘要】:目的:觀察肝缺血再灌注初期控制性降壓的干預(yù)作用,是否對(duì)大鼠腦內(nèi)過(guò)氧化物酶體增殖物激活受體γ(PPAR-γ)mRNA表達(dá)產(chǎn)生影響,并探討其機(jī)制。方法:清潔級(jí)雄性SD大鼠30只,隨機(jī)分為3組(n=10只):假手術(shù)組,肝缺血再灌注組,控制性降壓組。假手術(shù)組(S組)僅游離肝門,不阻斷肝血管;肝缺血再灌注組(I/R組)阻斷肝門60min后再灌注24h,建立缺血再灌注損傷模型;控制性降壓組(CH組)阻斷肝門60min,在再灌注初期股靜脈給予硝普鈉和艾司洛爾的混合液,控制性降壓至70mmHg,維持目標(biāo)血壓灌注10min,停藥至血壓回升后再灌注24h。24h后處死大鼠,斷頸取腦組織,左側(cè)腦組織制作石蠟切片用于病理觀察,右側(cè)腦組織采用比色法測(cè)定超氧化物歧化酶(SOD)活性及丙二醛(MDA)含量,RT-PCR檢測(cè)腦組織PPAR-γmRNA的相對(duì)表達(dá)。結(jié)果:光鏡下S組腦組織結(jié)構(gòu)未見異常;CH組腦組織損傷程度較I/R組減輕。與S組比較,I/R組和CH組的MDA含量和PPAR-γmRNA的相對(duì)表達(dá)顯著增高(P0.05),SOD活性顯著降低(P0.05);CH組腦組織中的MDA含量和PPAR-γmRNA的相對(duì)表達(dá)低于I/R組(P0.05),而SOD活性顯著升高(P0.05)。結(jié)論:再灌注初期控制性降壓可減輕大鼠肝缺血再灌注腦損傷有一定的腦保護(hù)作用,機(jī)制可能與降低MDA含量,升高SOD活性,下調(diào)PPAR-γmRNA表達(dá)等有關(guān)。
[Abstract]:Aim: to observe the effect of controlled hypotension on the expression of peroxisome proliferator activated receptor 緯 (PPAR- 緯) mRNA in rat brain at the early stage of hepatic ischemia reperfusion, and to explore its mechanism. Methods: thirty clean-grade male SD rats were randomly divided into 3 groups (n = 10): sham-operated group, hepatic ischemia-reperfusion group and controlled hypotension group. The model of ischemia-reperfusion injury was established in the sham operation group (S group), and the hepatic blood vessels were not blocked in the sham operation group (S group), and the hepatic ischemia-reperfusion injury model was established in the hepatic ischemia-reperfusion group (I / R group) after 24 hours of reperfusion after the hepatic portal 60min was blocked. In the controlled hypotension group (CH group), the hepatic portal was blocked for 60 minutes, and the femoral vein was given a mixture of sodium nitroprusside and esmolol at the early stage of reperfusion. The hypotension was controlled to 70 mm Hg, and the target blood pressure was maintained for 10 minutes. The rats were killed after stopping medication until the blood pressure rose and then 24h.24h was perfused, then the brain tissue was severed, and the left brain tissue was made into paraffin sections for pathological observation. The activity of superoxide dismutase (SOD) and the content of malondialdehyde (MDA) in right brain tissue were measured by colorimetry, and the relative expression of PPAR- 緯 mRNA in brain tissue was detected by RT-PCR. Results: the structure of brain tissue in S group was not abnormal under light microscope, and the degree of brain damage in CH group was less than that in I R group. Compared with S group, the content of MDA and the relative expression of PPAR- 緯 mRNA in I / R group and CH group were significantly higher than those in S group (P0.05), SOD activity was significantly lower than that in S group (P0.05); The content of MDA and the relative expression of PPAR- 緯 mRNA in brain tissue of CH group were lower than those of I / R group (P0.05), while the activity of SOD was significantly increased (P0.05). Conclusion: at the early stage of reperfusion, the controlled hypotension can alleviate the cerebral injury induced by hepatic ischemia and reperfusion in rats. The mechanism may be related to the decrease of MDA content, the increase of SOD activity and the down regulation of PPAR- 緯 mRNA expression.
