金錢(qián)魚(yú)DMRT1和GSDF在性別決定與分化中的功能初步研究

  魚(yú)類性別控制研究是現(xiàn)代水產(chǎn)養(yǎng)殖的熱點(diǎn)。金錢(qián)魚(yú)(Scatophagus argus,Linnaeus,1766)是一種廣鹽性魚(yú)類,可作觀賞魚(yú)和食用魚(yú),在養(yǎng)殖群體中性比為1:1。金錢(qián)魚(yú)雌魚(yú)的生長(zhǎng)比雄魚(yú)快,個(gè)體大,有更好經(jīng)濟(jì)價(jià)值。因此,克隆性別決定主效基因(master sex-determination gene,SD)將有助于金錢(qián)魚(yú)的性別控制。本研究旨在研究金錢(qián)魚(yú)的性別決定和分化機(jī)制,并為以后的單性(全雌魚(yú))金錢(qián)魚(yú)培育提供理論基礎(chǔ)。下面為本研究的主要發(fā)現(xiàn):1、金錢(qián)魚(yú)性別特異性標(biāo)記的開(kāi)發(fā)和性別決定候選基因的克隆本研究克隆了金錢(qián)魚(yú)Dmrt1(Dmrt1和Dmrt1b)和Dmrt3(Dmrt3和Dmrt3△-Y)的兩個(gè)亞型。本研究開(kāi)發(fā)了兩個(gè)位于金錢(qián)魚(yú)Dmrt3和Dmrt1上的性別連鎖分子標(biāo)記,分別命名為Dmrt3-Marker-F/R和Dmrt1-Marker-F/R。Dmrt3-Marker-F/R擴(kuò)增的片段包含能導(dǎo)致截短型Dmrt3(Dmrt3△-Y)c DNA提前編碼終止的基因組序列。Dmrt3-Marker-F/R擴(kuò)增片段的151和166 bp單核苷酸多態(tài)性(SNP)顯示雌性個(gè)體均為C...
