Development and Validation of KASP Markers for Wheat Breedin

功能標記(Functional markers,FMs)在作物育種過程中具有正要價值,功能標記的高通量基因型檢測為分子標記輔助選擇(Marker-assisted selection,MAS)在育種過程中的應用提供了很好的機會。在本研究中,我們開發(fā)并鑒定了一批與普通小麥適應性、產量、品質、生物和非生物脅迫抗性相關的KASP(Kompetitive Allele Specific PCR)標記,并對152個來自于本研究和公共數據庫的KASP標記在實際應用中進行可靠性鑒定。我們在以下三個方面對152個KASP標記中的70個進行可靠性驗證:1)在23個不同的小麥株系中對開發(fā)的KASP標記和基于凝膠的傳統(tǒng)PCR標記進行比較,2)利用來自中國和其他13個國家的300份品種的表型數據來驗證開發(fā)標記等位基因信息的有效性,3)在四個小麥分離群體中對開發(fā)的KASP標記進行鑒定。研究結果表明所有的KASP標記都與栽培品種和雙親群體中的相關表型有顯著的關聯,從而揭示了 KASP標記在小麥育種中潛在的應用前景。本研究表明KASP標記的檢測效率是基于凝膠的傳統(tǒng)PCR標記的45倍。目前,KASP標記已發(fā)展為一種單...
