甘蔗線條花葉病毒RT-LAMP檢測(cè)方法的建立及應(yīng)用(英文)

【目的】建立甘蔗線條花葉病毒(Sugarcane streak mosaic virus,SCSMV)的RT-LAMP檢測(cè)方法,為SCSMV的檢測(cè)提供技術(shù)支持!痉椒ā扛鶕(jù)甘蔗線條花葉病毒外殼蛋白基因的保守序列設(shè)計(jì)特異引物,優(yōu)化RT-LAMP的反應(yīng)條件,對(duì)比分析RT-LAMP的檢測(cè)靈敏度和特異性;并在RT-LAMP反應(yīng)產(chǎn)物中加入SYBR GREEN I進(jìn)行可視化檢測(cè),方便快速判定結(jié)果!窘Y(jié)果】成功建立了一種用于檢測(cè)SCSMV的RT-LAMP方法,可在60 ~ 63 ℃下60 min完成對(duì)SCSMV的檢測(cè),且具有較高的靈敏度,比傳統(tǒng)RT-PCR高100 倍,最低檢測(cè)RNA濃度為167.9×10-5 ng。此外,該方法具有較高的特異性,可特異檢測(cè)SCSMV。【結(jié)論】建立了一種靈敏、簡(jiǎn)便、快速的SCSMV RT-LAMP檢測(cè)方法并用于田間甘蔗樣品的檢測(cè)。
