【摘要】：瓜氨酸化是指精氨酸在鈣離子存在的情況下,由肽酰基精氨酸脫亞胺酶介導(dǎo)引起的一種翻譯后修飾。蛋白質(zhì)瓜氨酸化修飾的上調(diào)參與體內(nèi)多種疾病的發(fā)病機(jī)制,并具有重要的病理生理學(xué)意義。組織細(xì)胞中瓜氨酸化位點(diǎn)的鑒定依然迫切需要多種具有更高敏感性和特異性的定量和高通量的檢測(cè)方法,隨著化學(xué)衍生技術(shù)和質(zhì)譜技術(shù)的發(fā)展,新的快速精準(zhǔn)的蛋白質(zhì)組學(xué)技術(shù)依然具有較好的發(fā)展和應(yīng)用前景。文章主要通過(guò)介紹顯色劑分析、免疫識(shí)別、質(zhì)譜分析等多種瓜氨酸化多肽的檢測(cè)方法,以期為復(fù)雜生物樣本中瓜氨酸位點(diǎn)的鑒定提供新的思路和方向。
[Abstract]:Citrullination is a posttranslational modification of arginine mediated by peptidyl arginine deiminase in the presence of calcium ions. The upregulation of protein amino acid modification is involved in the pathogenesis of many diseases in vivo and has important pathophysiological significance. The identification of citrullinated sites in tissue cells is still in urgent need of more sensitive and specific quantitative and high-throughput detection methods, with the development of chemical derivation and mass spectrometry. New rapid and accurate proteomics technology still has a good prospect of development and application. In this paper, the methods of detection of citrullinated polypeptides such as colorimetric analysis, immune recognition and mass spectrometry are introduced in order to provide a new idea and direction for the identification of citrullinic acid sites in complex biological samples.
