【摘要】：目的比較TRIzol法與磁珠法對(duì)丙型肝炎病毒(HCV)RNA定量檢測(cè)結(jié)果的影響。方法收集117例丙型肝炎病毒感染陽(yáng)性患者的血清及其基因分型信息。分別采用TRIzol法與磁珠法提取患者血清樣本的HCV RNA,qPCR檢測(cè)提取HCV RNA的病毒載量,分析兩種提取方法所得HCV RNA檢測(cè)結(jié)果的差異。結(jié)果 qPCR結(jié)果顯示TRIzol法和磁珠法具有良好的線性相關(guān)性:y=0.978x+0.063(R2=0.973)。Bland-Altman統(tǒng)計(jì)分析顯示TRIzol法提取的HCV RNA病毒載量對(duì)數(shù)值的平均值略低于磁珠法,但差異無統(tǒng)計(jì)學(xué)意義(P0.05)�；蛐头治鼋Y(jié)果顯示1a、1b、2a、3a和6a基因型中兩種提取方法差異均無統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論 TRIzol法提取HCV RNA的定量檢測(cè)結(jié)果和磁珠法相當(dāng),且成本低廉,可在國(guó)內(nèi)廣泛用于試劑盒的開發(fā)和HCV RNA臨床檢測(cè)。
[Abstract]:Objective to compare the effect of TRIzol method and magnetic beads method on the quantitative detection of hepatitis C virus (HCV) RNA. Methods the serum and genotyping information of 117 patients with hepatitis C virus infection were collected and extracted by TRIzol method and magnetic beads method respectively. The viral load of HCV RNA extracted from patients' serum samples was detected by HCV RNA-qPCR. Results the qPCR results showed that there was a good linear correlation between the TRIzol method and the magnetic bead method. The statistical analysis showed that the average value of the HCV RNA virus load extracted by TRIzol method was slightly lower than that by the magnetic beads method, and the statistical analysis showed that there was a good linear correlation between the TRIzol method and the magnetic bead method, and the statistical analysis showed that the HCV RNA virus load extracted by TRIzol method was slightly lower than that by the magnetic bead method. The results of genotype analysis showed that there was no significant difference between the two methods for extracting HCV RNA from 1a and 6a genotypes. Conclusion the quantitative detection results of HCV RNA by TRIzol method are comparable to those by magnetic beads method and the cost is low. It can be widely used in the development of kit and clinical detection of HCV RNA in China.
