反相高效液相色譜法測(cè)定濕地根際土壤11種有機(jī)酸

【摘要】:研究了反相高效液相色譜法同時(shí)測(cè)定濕地植物根際土壤溶液中11種低分子有機(jī)酸的優(yōu)化方法。探索了柱溫、波長(zhǎng)、流速、流動(dòng)相中的pH值以及有機(jī)改良劑對(duì)檢測(cè)效果的影響,并進(jìn)一步驗(yàn)證方法的精密度、加標(biāo)回收率、檢測(cè)限等指標(biāo)。結(jié)果表明,采用Waters Atlantis T3 C18(4.6×250 mm,5μm)色譜柱,在柱溫為35℃,紫外檢測(cè)波長(zhǎng)210 nm,流動(dòng)相為磷酸鹽緩沖液(0.01 mol/L,pH 2.5)與乙腈(ACN)混合溶液,在流速0.7 m L/min時(shí),進(jìn)行梯度洗脫分離。該方法加標(biāo)回收率在95.6%~103.9%,相對(duì)標(biāo)準(zhǔn)偏差在0.3%~5.2%,各酸峰面積與質(zhì)量濃度線性關(guān)系良好(R2≥0.999)。該方法成功用于介質(zhì)復(fù)雜的環(huán)境樣品中低分子有機(jī)酸的檢測(cè),2014年4月采集的2種濕地挺水植物根際與非根際土壤溶液檢測(cè)主要存在草酸與丙二酸。蘆葦土壤溶液中草酸總計(jì)15.83 mg/L,丙二酸總計(jì)18.10 mg/L;扁稈呰草土壤溶液中草酸17.90 mg/L,丙二酸總計(jì)17.05 mg/L。2種植物中,根際土壤溶液中丙二酸含量均高于非根際土壤溶液。
