甜櫻桃根際IAA產(chǎn)生菌的篩

【摘要】:為了給甜櫻桃微生物肥料的研制和生產(chǎn)提供優(yōu)良菌株資源,本文從馬哈利櫻桃根際土壤中分離獲得52株細(xì)菌,通過(guò)保綠法和蘿卜子葉增重法進(jìn)行初篩,篩選具有促生作用的菌株,進(jìn)一步進(jìn)行IAA產(chǎn)生菌的篩選,并經(jīng)形態(tài)觀察、生理生化特征及16SrDNA序列分析明確分類地位,利用分光光度法測(cè)定產(chǎn)IAA的能力,并對(duì)最佳碳氮源、產(chǎn)素條件進(jìn)行了篩選。結(jié)果表明,52株菌中B20產(chǎn)IAA能力最強(qiáng),經(jīng)鑒定為阿氏芽孢桿菌(Bacillus aryabhattai),其最佳產(chǎn)素的培養(yǎng)條件為:采用30%的裝液量在溫度為30℃、轉(zhuǎn)速為150r·min-1的條件下培養(yǎng)44h,最佳碳、氮源分別為果糖和酵母粉。研究發(fā)現(xiàn)菌株B20具有IAA產(chǎn)生能力,具有作為功能微生物肥料研發(fā)的潛力,可進(jìn)一步對(duì)其進(jìn)行開(kāi)發(fā)研究。
[Abstract]:In order to provide excellent strain resources for the development and production of sweet cherry microbial fertilizer, 52 strains of bacteria were isolated from the rhizosphere soil of Mahari cherry and screened by green preserving method and radish cotyledon weight gain method. Further screening of IAA producing strains was carried out, and the taxonomic status was determined by morphological observation, physiological and biochemical characteristics and 16SrDNA sequence analysis. The ability of producing IAA was determined by spectrophotometry, and the optimum carbon and nitrogen sources were analyzed. The production conditions were screened. The results showed that B20 had the strongest ability to produce IAA among 52 strains.
