新蚜蟲癘霉菌絲和孢子階段的轉錄組學分析

【摘要】:新蚜蟲癘霉(Pandora neoaphidis)是蟲霉門(Entomophthoromycota)中極具代表性的蚜蟲;圆≡婢,分布廣泛,可侵染多種蚜蟲。適宜條件下可在較短的時期內形成流行病,在蚜蟲自然防治中起主導作用。為了系統(tǒng)分析了兩個階段的基因表達和功能相關的轉錄組信息,本研究通過二代測序平臺Illumina Hiseq 2500對新蚜蟲癘霉的兩種發(fā)育形態(tài)(菌絲和分生孢子)進行轉錄組測序。得到90 639個不含N(模糊堿基)的組裝片段(contigs)和49 138條單基因簇(unigenes)。49 138條單基因簇經基因本體論(Gene Ontology,GO)功能顯著性富集分類后歸到47種功能分支中,相關的基因功能主要歸類至分子功能、細胞組分和生化過程三大類中。根據(jù)每百萬reads中來自于某基因每千堿基長度的reads數(shù)(reads per kilobase per million of mapped reads,RPKM)的表達量算法一共得到3 555個差異表達的基因,其中上調的基因有2 294個,下調的基因有1 261個。利用京都基因及基因組百科全書(Kyoto Encyclopedia of Genes and Genomes,KEGG)對差異表達基因進行途徑顯著性富集分析后發(fā)現(xiàn)與之相關的代謝路徑有22條,其中糖胺聚糖降解和類胡蘿卜素生物合成兩條途徑顯著上調。本研究為進一步挖掘和分析與新蚜蟲癘霉生長發(fā)育相關的功能基因提供了理論依據(jù)。
[Abstract]:(Pandora neoaphidis) is a representative aphids specific to venereal fungi in (Entomophthoromycota). It is widely distributed and can infect many kinds of aphids. Under suitable conditions, epidemic diseases can be formed in a short period of time and play a leading role in the natural control of aphids. In order to systematically analyze the transcriptional information related to gene expression and function in two stages, the second generation sequencing platform Illumina Hiseq 2500 was used to sequence the two developmental forms (hyphae and conidium) of Pandora neoaphids. 90,639 (contigs) fragments without N (fuzzy base) and 49,138 single gene clusters (unigenes). 49,138 single gene clusters were classified into 47 functional branches by functional enrichment and classification of Gene Ontology,GO. The related gene functions are classified into three categories: molecular function, cellular component and biochemical process. A total of 3 555 differentially expressed genes were obtained according to the algorithm of reads number (reads per kilobase per million of mapped reads,RPKM per million reads, in which 2 294 genes were up-regulated and 1 261 genes were down-regulated. Significant enrichment of differentially expressed genes by using Kyoto gene and genome encyclopedia (Kyoto Encyclopedia of Genes and Genomes,KEGG showed that there were 22 related metabolic pathways. Glycosaminoglycan degradation and carotenoid biosynthesis were significantly up-regulated. This study provides a theoretical basis for further research and analysis of functional genes related to the growth and development of Pandora neoaphids.
