石柱黃連根腐病根際土壤細(xì)菌微生態(tài)研究

【摘要】:該研究應(yīng)用Illumina Hiseq 2500高通量測序平臺,分析了未種植黃連土壤、黃連健株及病株根際土壤的細(xì)菌種群豐富度及多樣性變化,并結(jié)合土壤養(yǎng)分及酶活變化檢測,對黃連根腐病產(chǎn)生的機(jī)制進(jìn)行了深入探討。高通量測序顯示,栽培黃連造成了土壤細(xì)菌種群的豐富度的顯著降低(P0.05)和細(xì)菌種群多樣性的降低;罹患根腐病的黃連植株根際土壤細(xì)菌的種群豐富度要顯著低于未種植和健康黃連土(P0.05),種群多樣性要顯著低于未種植黃連土樣(P0.05),但與健株土樣之間差異不顯著。對土壤的養(yǎng)分及酶活測定結(jié)果表明,栽培黃連造成了土壤pH、有效磷、脲酶活性的顯著降低,蔗糖酶活性的顯著升高(P0.05);根腐病土樣中有機(jī)碳的含量、堿解氮、過氧化氫酶和脲酶活性顯著低于健康土樣,速效磷和速效鉀含量顯著高于健康土樣(P0.05)。綜合分析表明,栽培黃連造成了土壤中細(xì)菌種群豐富度和多樣性的降低;病株土樣中,細(xì)菌種群豐富度的顯著降低和種群多樣性的降低,有可能是導(dǎo)致黃連根腐病產(chǎn)生的一個重要原因,此外,土壤有機(jī)碳和過氧化氫酶活性的顯著減低,也許是導(dǎo)致黃連根腐病發(fā)生的一個誘因。
[Abstract]:In this study, Illumina Hiseq 2500 high-throughput sequencing platform was used to analyze the variation of bacterial population richness and diversity in the rhizosphere soil of non-cultivated Coptis chinensis, Rhizosphere of Coptis chinensis and diseased plants. The mechanism of root rot of Coptis chinensis was studied with soil nutrient and enzyme activity detection. High throughput sequencing showed that the root rot of Coptis chinensis was caused by Rhizoma Coptidis. Cultivated Rhizoma Coptidis caused a significant decrease in soil bacterial population richness (P0.05) and bacterial population diversity. The population richness of rhizosphere bacteria in rhizosphere soil of Coptis chinensis infected with root rot was significantly lower than that of non-cultivated and healthy Coptis chinensis P0.05N, and the population diversity was significantly lower than that of non-cultivated soil samples of Coptis chinensis (P0.05). But there was no significant difference between soil samples and healthy soil samples. The results of soil nutrient and enzyme activity test showed that the soil pH, available phosphorus and urease activity of cultivated Coptis chinensis were significantly decreased. The activity of sucrase increased significantly (P 0.05). The contents of organic carbon, alkali-hydrolyzed nitrogen, catalase and urease in root rot soil samples were significantly lower than those in healthy soil samples, and the contents of available phosphorus and available potassium were significantly higher than that of healthy soil samples P0.050.The comprehensive analysis showed that the organic carbon content in soil samples was significantly higher than that in healthy soil samples. Cultivation of Coptis chinensis resulted in the decrease of bacterial population richness and diversity in soil. The significant decrease of bacterial population richness and the decrease of population diversity in soil samples of diseased plants may be an important cause of root rot of Coptis chinensis. A significant decrease in soil organic carbon and catalase activity may be a contributing factor to root rot of Coptis chinensis.
